摘 要:目的明确海洋生物来源的鱼I型胶原(marine type I collagen,MCI)对巨噬细胞生物学功能的影响,探讨其转化前景。方法诱导人源巨噬细胞,体外给予不同浓度MCI刺激,CCK8法观察MCI对巨噬细胞活性的影响;Transwell实验观察MCI对单核/巨噬细胞细胞趋化能力的影响。免疫荧光实验和RT-qPCR观察MCI对巨噬细胞表型的影响。转录组测序观察MCI对巨噬细胞生物学功能的影响。结果MCI在1mg/ml和3mg/ml的浓度下,在体外不会对巨噬细胞的细胞活性、单核/巨噬细胞的趋化功能及巨噬细胞炎性表型产生显著影响。进一步转录组测序及验证发现,MCI可以显著提高巨噬细胞CXCL13的表达。结论鱼I型胶原可通过免疫调控促进巨噬细胞表达CXCL13,该现象可能利于骨缺损修复,提示该鱼I型胶原有一定的应用前景。Objective Marinetype I collagen(MCI)has the most extensive sources in the all of the marine collagen.Herein,we aimed to explore the biological effects of MCI on the macrophages,and to demonstrate the translational values of MCI in the medical area.Methods We managed to construct a MCI-macrophage model,and CCK8 assays were performed to observe the activities of macrophages with the treatment of MCI.Transwell assays were used to observe the chemotaxis of monocytes/macrophages under the effects of MCI.Immunofluorescence and RT-qPCR were performed to demonstrate the phenotype alterations of macrophages.Besides,transcriptome sequencing was performed to discover the biological changes of macrophages under the treatment of MCI.Results The activities of macrophages and the migration abilities of monocytes/macrophages were not obviously changed under the treatment of MCI(concentration:1mg/ml and 3mg/ml).MCI could not cause the inflammatory phenotypes of macrophages.Transcriptome sequencing and subsequent validation indicated MCI could significantly increase the expression of CXCL13.Conclusions MCI is the most available marine collagen.In this study,we demonstrated MCI could increase the expression of CXCL13 in macrophages,indicating the translational values of MCI in the regeneration of bone defects.
