摘 要:目的采用高效液相色谱法建立朝鲜淫羊藿药材指纹图谱,并通过化学模式识别和多元统计分析,探析表征不同产地朝鲜淫羊藿质量差异的标志物。方法采用高效液相色谱法建立9个产地朝鲜淫羊藿药材指纹图谱,通过对照品比对和高效液相色谱-四级杆飞行时间串联质谱技术对共有峰进行指认,并利用相似度评价结合聚类分析、主成分分析、正交偏最小二乘法-判别分析等统计分析方法探析不同产地朝鲜淫羊藿药材的差异性成分。结果9个产地朝鲜淫羊藿指纹图谱共标定11个共有峰,与对照指纹图谱的相似度均在0.977以上。聚类分析将9个产地朝鲜淫羊藿样品分为3类,主成分分析和正交偏最小二乘法-判别分析结果与聚类分析一致,并筛选得到6个产地差异标志物,分别为色谱峰1(金丝桃苷)、色谱峰9(朝鲜淫羊藿苷B)、色谱峰6(淫羊藿苷)、色谱峰4(朝藿定B)、色谱峰7[Anhydroicaritin-3-O-rhamnoside(1-2)-furanacid-7-O-glucoside]和色谱峰11。结论朝鲜淫羊藿在不同产地之间存在一定的质量差异,建立的朝鲜淫羊藿指纹图谱可为药材质量评价提供依据。Objective The fingerprint of Chaoxian Yinyanghuo(Epimedium koreanum Nakai)was established by HPLC,and the markers characterizing the quality differences of Chaoxian Yinyanghuo(Epimedium koreanum Nakai)from different producing areas were analyzed by chemical pattern recognition and multivariate statistical analysis.Method The fingerprints of Chaoxian Yinyanghuo(Epimedium koreanum Nakai)from 9 producing areas were established by HPLC,the common peaks were identified by comparison of reference substances and HPLC-Q-TOF-MS.The differential components of Chaoxian Yinyanghuo(Epimedium koreanum Nakai)from different producing areas were analyzed by similarity evaluation,cluster analysis,principal component analysis,orthogonal partial least squares discriminant analysis and other statistical analysis methods.Results The fingerprints of Chaoxian Yinyanghuo(Epimedium koreanum Nakai)from 9 producing areas were calibrated with 11 common peaks,and the similarity with the control fingerprints was more than 0.977.The samples of Chaoxian Yinyanghuo(Epimedium koreanum Nakai)from 9 producing areas were divided into 3 categories by cluster analysis.The results of principal component analysis and orthogonal partial least squares discriminant analysis were consistent with cluster analysis,and 6 markers of difference in producing areas were screened,which were peak 1(hypericin),peak 9(korepimedoside B),peak 6(icariin),peak 4(epimedin B),peak 7(anhydroicaritin-3-O-rhamnoside(1-2)-furanacid-7-O-glucoside)and peak 11.Conclution There are some quality differences among different producing areas of Chaoxian Yinyanghuo(Epimedium koreanum Nakai)and the established fingerprint of Chaoxian Yinyanghuo(Epimedium koreanum Nakai)can provide a basis for the quality evaluation of medicinal materials.
