摘 要:目的探讨人参皂苷Rg1通过LncRNA MALAT1通路对脊髓损伤大鼠神经再生修复的影响。方法30只大鼠随机分为Rg1治疗组、假手术组、脊髓损伤对照组。将成年雌性鼠随机分为假手术组,脊髓损伤对照组和Rg1治疗组,建立大鼠脊髓损伤模型。Rg1治疗组大鼠每天腹腔注射Rg1溶液(100 mg/kg),脊髓损伤对照组和假手术组每天腹腔注射等量生理盐水。采用Basso-Beattie-Bresnahan(BBB)评分评估大鼠脊髓损伤后的运动恢复情况。连续治疗28 d后处死老鼠。取脊髓组织,通过Western blot检测细胞黏附分子层粘连蛋白(Laminin,LN)、纤维连接蛋白(Fibronectin,FN)以及神经营养因子胶质细胞源性神经营养因子(glialcellline-derivedneurotrophicfactor,GDNF)、神经生长因子(nerve growth factor,NGF)的表达。实时荧光定量PCR测LncRNA MALAT1,采用免疫组化分析LAMC1的表达水平。结果BBB评分显示,Rg1治疗组BBB评分显著优于假手术组、脊髓损伤对照(P<0.05)。免疫组化及免疫荧光显示:Rg1治疗组LN、FN、GDNF、NGF、LAMC1表达与对照组相比显著升高。结论Rg1能够上调LncRNA MALAT1表达,进一步调控LAMC1而促进大鼠脊髓损伤修复。Objective To explore the effects of ginsenoside RG1 through LncRNA MALAT1 pathway on spinal cord injury to the regeneration of rats nerve regeneration.Methods Thirty rats were randomly divided into RG1 treatment group,sham operation group and spinal cord injury control group.Adult female rat was randomly divided into sham operation group,spinal cord injury control group and RG1 treatment group,and a rat spinal cord injury model was established.The rats in Rg1 treatment group were intraperitoneally injected with Rg1 solution(100 mg/kg)every day,and the rats in spinal cord injury control group and sham operation group were intraperitoneally injected with the same amount of normal saline every day.Basso Beattie Bresnahan(BBB)score was used to evaluate the motor recovery after spinal cord injury in rats.The rats were killed after 28 days of continuous treatment.The spinal cord tissue was taken out and the expressions of laminin(LN),fibronectin(FN),glial cell derived neurotrophic factor(GDNF)and nerve growth factor(NGF)were detected by Western blot.Real time fluorescent quantitative PCR was used to measure LncRNA MALAT1,and immunohistochemistry was used to analyze the expression level of LAMC1.Results The BBB score of the RG1 treatment group was significantly better than that of the sham operation group and spinal cord injury control group(P<0.05).Immunization and immune fluorescent displayed RG1 the expressions of LN,FN,GDNF,NGF and LAMC1 in the treatment group were significantly increased compared with those of the control group.Conclusion RG1 can increase the expressions of LNCRNA MALAT1 and further regulate LAMC1 to promote the repair of spinal cord injury in rats.
