摘  要:目的探究健脾祛痰方通过过氧化物酶体增殖物激活受体γ(peroxisome proliferatoractivated receptor gamma,PPARγ)-肝X受体α(liver Xreceptor alpha,LXRα)-三磷酸腺苷结合盒转运体A1(ATP binding cassette transporter A1,ABCA1)信号通路干预血脂异常的作用机制。方法采用随机分组法将18只SPF级雄性SD大鼠分为造模组(n=12)和对照组(n=6)。造模组饲喂高脂饲料,对照组饲喂普通饲料。造模成功的大鼠随机分为模型组(n=6)和健脾祛痰方组(n=6)。健脾祛痰方组予中药健脾祛痰方颗粒剂灌胃,对照组、模型组予生理盐水灌胃,干预4周。4周后检测各组大鼠血清胆固醇(total cholesterol,TC)、甘油三酯(triglyceride,TG)、高密度脂蛋白胆固醇(high-density lipoprotein cholesterol,HDL-C)、低密度脂蛋白胆固醇(low-density lipoprotein cholesterol,LDL-C)的水平;油红o染色观察各组大鼠肝脏脂质沉积变化;实时荧光定量PCR技术及免疫印迹实验检测各组大鼠肝脏组织中PPARγ、LXRɑ、ABCA1、B族Ⅰ型清道夫受体(scavenger receptor class B type I,SR-B1)、胆固醇7α-羟化酶(cholesterol 7α-hydroxylase,CYP7A1)mRNA及蛋白表达。结果与对照组相比,模型组大鼠肝脏出现脂质沉积;血清TC、TG、LDL-C水平升高(P<0.05)、HDL-C水平有改变,统计学统计显示变化不显著(P>0.05);肝脏PPARγ、LXRα、ABCA1、SR-B1、CYP7A1 mRNA及蛋白表达下降(P<0.05)。对比模型组,健脾祛痰方组大鼠肝脏脂质沉积面积减少;肝脏PPARγ、LXRα、ABCA1、SR-B1、CYP7A1 mRNA及蛋白表达升高(P<0.05)。干预前后比较,干预后健脾祛痰方组大鼠血清TC、TG、LDL-C水平下降(P<0.05),HDL-C水平升高,差异无统计学意义(P>0.05)。结论中药健脾祛痰方可能通过调控PPARγ-LXRα-ABCA1信号通路改善高脂饮食诱导的血脂异常。Objective To explore the mechanism of Jianpi Qutan Recipe(健脾祛痰方)on dyslipidemia through peroxisome proliferator activated receptor gamma(PPARγ)/liver X receptor alpha(LXRα)/ATP binding cassette transporter A1(ABCA1)signaling pathway.Methods Eighteen SPF male SD rats were randomly divided into model group(n=12)and control group(n=6).The modeling group was fed with high-fat feed,and the control group was fed with ordinary feed.Rats with successful modeling were randomly divided into model group(n=6)and Jianpi Qutan Recipe group(n=6).The Jianpi Qutan Recipe group was given Jianpi Qutan Recipe Granules by gavage,while the control group and the model group were given normal saline by gavage for 4 weeks of intervention.After 4 weeks,the serum levels of total cholesterol(TC),triglyceride(TG),high density lipoprotein cholesterol(HDL-C)and low density lipoprotein cholesterol(LDL-C)were detected in the rats in each group.The lipid deposition changes in the liver of the rats in each group were observed by Oil Red O staining.The expressions of PPARγ,LXRɑ,ABCA1,Scavenger receptor class B type I(SR-B1),cholesterol 7α-hydroxylase(CYP7A1)mRNA and protein in rats liver tissue were tested by Real time Fluorescence Quantitative PCR Technique and Western blot.Results Compared with those of the control group,lipid deposition occurred in the liver of the model group.The serum levels of TC,TG and LDL-C increased(P<0.05),and HDL-C levels have changed,but statistics showed no significant change of statistics(P>0.05).The expressions of PPARγ,LXRα,ABCA1,SR-B1 and CYP7A1 mRNA and protein decreased(P<0.05).Compared with those of the model group,the liver lipid deposition area of the rats in the Jianpi Qutan Recipe group decreased.The mRNA and protein expressions of PPARγ,LXRα,ABCA1,SR-B1 and CYP7A1 in the liver increased(P<0.05).The serum levels of TC,TG and LDL-C decreased in the Jianpi Qutan Recipe group after the intervention(P<0.05),and HDL-C levels increased,but statistics showed no significant change of statistics(P>
