摘  要:目的探讨中成药醒脑静对控制性降压大鼠脑组织海马区神经元凋亡的影响,研究其对预防和治疗控制性降压大鼠复压后脑缺血再灌注损伤的可能机制。方法选取SPF级SD大鼠40只,按照随机数字表法随机分为4组:空白对照组、控制性降压组、醒脑静低剂量治疗组、醒脑静高剂量治疗组。采用翻正实验、第1次舔足时间、舔足次数及运动斜板实验进行大鼠行为测试和神经行为功能评价,应用免疫组织化学法及Western-blot法检测SD大鼠海马组织中天冬半胱氨酸蛋白酶-1(Caspase-1)、天冬半胱氨酸蛋白酶-3(Caspase-3)及HSP70的表达水平。结果与空白对照组比较,控制性降压组运动、感觉功能显著降低(P<0.05,P<0.01);与控制性降压组比较,醒脑静高、低剂量治疗组运动、感觉功能显著提高(P<0.01);与醒脑静低剂量治疗组比较,醒脑静高剂量治疗组运动、感觉功能显著提高(P<0.01)。与空白对照组比较,控制性降压组HSP70、Caspase-1、Caspase-3蛋白表达水平显著升高(P<0.01);与控制性降压组比较,醒脑静高、低剂量治疗组HSP70蛋白表达水平显著升高(P<0.01),Caspase-1、Caspase-3蛋白表达水平显著降低(P<0.01);与醒脑静低剂量治疗组比较,醒脑静高剂量治疗组HSP70蛋白表达水平显著升高(P<0.01),Caspase-1、Caspase-3蛋白表达水平显著降低(P<0.01)。免疫组织化学染色及结果显示,大鼠海马组织中HSP70阳性表达产物呈棕黄色颗粒状,Caspase-1、Caspase-3阳性表达产物呈棕褐色颗粒状,附着于神经元细胞膜和细胞质中。控制性降压组HSP70阳性表达产物高于空白对照组,醒脑静高剂量治疗组海马组织中HSP70阳性表达产物高于醒脑静低剂量治疗组。控制性降压组Caspase-1、Caspase-3阳性表达产物高于空白对照组,醒脑静高剂量治疗组海马组织中Caspase-1、Caspase-3阳性表达产物低于低剂量醒脑静治疗组。采用WB法测定醒脑静高剂量治疗�Objective To investigate the influence of the Chinese patent medicine Xingnaojing(醒脑静)on the apoptosis of neurons in the hippocampal region of rat brain tissue under controlled hypotension and the possible mechanism of its effect on the treatment of ischemia-reperfusion injury in the brain after recovery of blood pressure in rats under controlled hypotension.Methods Forty SD rats of SPF class were randomly divided into four groups:blank control group,controlled hypotensive group,Xingnaojing lowdose treatment group,and Xingnaojing high-dose treatment group.The rats were carried out the behavioral testing by behavioral testing and neurobehavioral function evaluation,using the righting reflex experiment,the inclined plane test,and recording the time and number of the first foot licking.The expression levels of apoptosis-related proteins Caspase-1,Caspase-3 and HSP70 in the hippocampal region were detected by immunohistochemistry and western-blot method.Results Compared with the blank control group,the motor and sensory functions in the controlled hypotension group were significantly decreased(P<0.05,P<0.01);Compared with the controlled hypotension group,the motor and sensory functions of the high-dose and low-dose Xingnaojing treatment groups were significantly improved(P<0.01);Compared with the low dose Xingnaojing treatment group,the high dose Xingnaojing treatment group significantly improved motor and sensory functions(P<0.01).Compared with the blank control group,the expression levels of HSP70,Caspase-1,and Caspase-3 proteins in the controlled hypotension group were significantly increased(P<0.01);Compared with the controlled hypotension group,the expression levels of HSP70 protein in the high-dose and low-dose Xingnaojing treatment groups were significantly increased(P<0.01),while the expression levels of Caspase-1 and Caspase-3 protein were significantly decreased(P<0.01);Compared with the low dose Xingnaojing treatment group,the high dose Xingnaojing treatment group significantly increased the level of
