摘 要:目的研究通过观察麻杏石甘汤对卵清蛋白(OVA)造模过敏性哮喘大鼠肺组织中辅助T淋巴细胞9(Th9)、辅助T淋巴细胞2(Th2)、辅助T淋巴细胞17(Th17)的调控作用,探讨麻杏石甘汤干预过敏性哮喘的可能机制。方法采用OVA复制过敏性哮喘大鼠模型,使用麻杏石甘汤以及对照药物孟鲁司特按分组给予相应干预后,采用ELISA法检测各组大鼠血清免疫球蛋白E(IgE)、白介素-4(IL-4)、转化生长因子-β(TGF-β)、白介素-9(IL-9)、白介素-5(IL-5)、白介素-17(IL-17)表达,采用Western-blot法检测大鼠肺组织中IL-4、TGF-β、IL-9表达,采用流式细胞术检测外周血淋巴细胞中Th2、Th9、Th17比例。结果(1)麻杏石甘汤组大鼠肺组织可见支气管黏膜慢性炎症较轻,杯状细胞、黏膜增生突起较轻,肺气肿及肺间质性炎症较轻,周围存在少数肥大细胞、嗜酸性粒细胞等炎症细胞浸润。(2)与空白对照组相比,其他各组大鼠血清中IgE、IL-4、TGF-β、IL-9、IL-5、IL-17表达均明显升高(P<0.05);与模型组相比,药物组大鼠血清中IgE、IL-4、TGF-β、IL-9、IL-5、IL-17表达均明显减低(P<0.05),西药对照组和麻杏石甘汤组组间比较差异无统计学意义(P>0.05)。(3)与空白对照组比较,其他各组大鼠肺组织IL-4、TGF-β、IL-9表达水平显著上调(P<0.05);与模型对照组比较,西药对照组、麻杏石甘汤组大鼠肺组织IL-4、TGF-β、IL-9表达水平显著下调(P<0.05);与西药对照组比较,麻杏石甘汤组大鼠肺组织IL-4、TGF-β、IL-9表达水平显著下调(P<0.05)。(4)与空白对照组比较,模型对照组、西药对照组中Th2、Th9、Th17比例显著提高(P<0.05),麻杏石甘汤组中Th2、Th17比例显著提高(P<0.05),但Th9比例比较差异无统计学意义(P>0.05);与模型对照组相比,西药对照组和麻杏石甘汤组中Th2、Th9、Th17比例降低(P<0.05);与西药对照组比较,麻杏石甘汤组大鼠外周血淋巴细胞中Th2、Th17比例组间比较差异�Objective Through observing Maxing Shigan Decoction(麻杏石甘汤)regulating effect on helper T lymphocytes 9(Th9),helper T lymphocytes 2(Th2),and helper T lymphocytes 17(Th17)in rat lung tissue of ovalbumin(OVA)molding allergic asthma,this study discussed the possible mechanism of Maxing Shigan Decoction on allergic asthma.Methods After adopting OVA to replicate allergic asthma rat model,Maxing Shigan Decoction and the control drug Montelukast were used for corresponding intervention in groups.ELISA method was used to detect the content of IgE,IL-4,TGF-β,IL-9,IL-5 and IL-17 in serum of rats.The expressions of IL-4,TGF-βand IL-9 in rat lung tissues were detected by Western-blot method.The percentages of Th2,Th9 and Th17 in peripheral blood lymphocytes were determined by flow cytometry.Results(1)Chronic inflammation of bronchial mucosa was mild,the proliferative processes of goblet cells were light,emphysema and pulmonary interstitial inflammation were mild,there were a few inflammatory cells such as mast cells and eosinophils around.(2)Compared with the blank control group,the expressions of IgE,IL-4,TGF-β,IL-9,IL-5 and IL-17 in serum of rats in other groups increased significantly(P<0.05);Compared with the model group,the expressions of serum IgE,IL-4,TGF-β,IL-9,IL-5 and IL-17 in the drug group decreased significantly(P<0.05).There was no significant difference between the Western medicine control group and Maxing Shigan Decoction group(P>0.05).(3)Compared with the control group,the expression levels of IL-4,TGF-β,IL-9 in lung tissue of the other groups were up-regulated(P<0.05);Compared with model control group,the expression levels of IL-4,TGF-β,IL-9 in lung tissue of Maxing Shigan Decoction group were down-regulated significantly(P<0.05);Compared with the control group,the expression levels of IL-4,TGF-β,IL-9 in lung tissue of Maxing Shigan Decoction group were significantly down-regulated(P<0.05).(4)Compared with the blank control group,the proportions of Th2,Th9,Th17 in the model control group were
