摘 要:目的探讨感染所致复发性流产(RSA)孕妇非编码核糖核苷酸(lncRNA)SOX2OT基因多态性,评估其对RSA的作用机制及影响。方法选择2018年11月-2021年11月天津市第五中心医院收治的因RSA入院孕妇62例为RSA组,选择同期处于孕早期(<7周)来院自愿要求终止妊娠孕妇62例为对照组。抽取外周静脉血标本,采用实时荧光定量聚合酶链式反应(RT-PCR)检测lncRNA SOX2OT基因rs9839776 C>T位点基因分布。结果两组孕妇年龄、入院时孕周、体质量指数、受孕方式和空腹血糖受损情况比较差异无统计学意义;两组lncRNA SOX2OT基因rs9839776 C>T位点实际频数和理论频数分布符合哈迪-温伯格平衡,具有群体代表性;RSA组lncRNA SOX2OT基因rs9839776 C>T位点TT基因型和T等位基因频率均高于对照组(P<0.05);RSA组不同基因型孕妇携带病原情况比较差异无统计学意义。结论lncRNA SOX2OT基因rs9839776 C>T位点TT基因型可能增加生殖道感染孕妇发生RSA的风险,但与感染病原无关。OBJECTIVE To explore gene polymorphisms of long non-coding ribonucleic acid(lncRNA)SOX20T In pregnant women with infection-induced recurrent spontaneous abortion(RSA)and evaluate the action mechanisms and effect on RSA.METHODS A total of 62 pregnant women who were hospitalized in Tianjin Fifth Central Hospital due to RSA from Nov 2018 to Nov 2021 were assigned as the RSA group,meanwhile,62 early pregnancy women(less than 7 weeks)who volunteered to terminate the pregnancy were chosen as the control group.The peripheral venous blood specimens were collected,and the genotypes at rs9839776 CT loci of lncRNA SOX2OT gene were detected by means of real-time fluorescent quantitative polymerase chain reaction(RT-PCR).RESULTS There were no significant differences in the age,gestational weeks at the admission,body mass index,conception mode and impaired fasting blood glucose between the two group.The actual frequency and theoretical frequency of rs9839776 CT loci of lncRNA SOX2OT gene were in line with the Hardy-Weinberg equilibrium and were representative of the population.The frequencies of TT genotype and T allele at rs9839776 CT loci of lncRNA SOX2OT gene were higher in the RSA group than in the control group(P<0.05).In the RSA group,there was no significant difference in the status of carrying with pathogens among the pregnant women with different genotypes.CONCLUSION The TT genotype at rs9839776 CT loci of lncRNA SOX2OT gene may increase the risk of RSA among the pregnant women with genital tract infection but are not associated with the infectious pathogens.Download(CAJ format)Download(PDF format)
