摘  要:目的探讨筋骨草水煎液对顺铂所致肾损伤的预防作用。方法取成年雄性昆明小鼠30只,随机分为对照组、顺铂组、筋骨草组,每组各10只。筋骨草组给予筋骨草水煎液灌胃,对照组和顺铂组给予生理盐水灌胃,每天1次,连续12 d;于实验第7天筋骨草水煎液灌胃2 h后,顺铂组和筋骨草组给予10 mg/kg顺铂一次性腹腔注射诱导肾损伤。处死小鼠,取肾脏,用HE染色观察肾组织病理学改变;采集血液,使用全自动生化分析仪测定BUN、Cr、CysC,ELISA法检测血清TNF-α、IL-1β;取肾组织,WST-1法检测SOD,TBA法检测MDA,微板法检测GSH;RT-qPCR法检测氧化应激(Nrf2、HO-1、NQO1)、凋亡(Bcl-2、Caspase-3、Bax)相关mRNA表达。结果与对照组比较,顺铂组肾小球体积增大,肾小管上皮细胞明显肿胀;血清BUN、Cr、CysC、TNF-α、IL-1β水平升高;肾组织MDA含量升高,SOD和GSH含量降低;肾组织Nrf2、HO-1、NQO1、Caspase-3、Bax mRNA表达上调,Bcl-2 mRNA表达下调。与顺铂组比较,筋骨草组肾组织病理损伤减轻;血清BUN、Cr、CysC、TNF-α、IL-1β水平降低;肾组织MDA含量下降、SOD、GSH含量升高,Nrf2、HO-1、NQO1、Bcl-2 mRNA表达升高,Caspase-3、Bax mRNA表达降低(P<0.05或<0.01)。结论筋骨草水煎液对顺铂所致肾损伤具有治疗作用,该作用可能是通过激活Nrf2信号通路、抑制NF-κB和Caspase-3信号通路进而抑制氧化应激、炎症反应和细胞凋亡实现的。Objective To investigate the protective effect of Ajuga decumbens Thunb aqueous decoction on renal in⁃jury mice induced by cisplatin.Methods Thirty adult male Kunming mice were randomly divided into the control group,cisplatin group and Ajuga decumbens Thunb group,with 10 mice in each group.Mice in the Ajuga decumbens Thunb group were given Ajuga decumbens Thunb aqueous decoction by intragastric administration,while mice in the con⁃trol group and cisplatin group were given normal saline through intragastric administration,once a day,for 12 days.On the 7th day of the experiment,renal injury was induced by intraperitoneal injection of 10 mg/kg cisplatin in the cisplatin group and the Ajuga decumbens Thunb group for 2 h after intragastatic administration of Ajuga decumbens Thunb aqueous decoction.The mice were sacrificed,the kidneys were collected,and the histopathological changes of the kidneys in the three groups were observed by HE staining.Blood was collected,BUN,Cr,and CysC were detected by the fully automat⁃ed biochemical analyzer,and the levels of TNF-αand IL-1βin serum were determined by ELISA.WST-1 method was used to detect SOD,TBA method to detect MDA,and microplate method to detect GSH.The mRNA expression levels of oxidative stress-related Nrf2,HO-1 and NQO1 and apoptosis-related Bcl-2,Caspase-3 and Bax were detected by RT-qP⁃CR.Results Compared with the control group,the glomeruli volume of mice in the cisplatin group was enlarged and the epithelial cells of renal tubules became significantly swollen,the concentrations of BUN,Cr,CysC,TNF-αand IL-1βin serum increased,MDA levels increased,SOD and GSH levels decreased,Nrf2,HO-1,NQO1,Caspase-3 and Bax expression levels were up-regulated while the Bcl-2 expression was down-regulated in renal tissues.Compared with the cispl⁃atin group,the pathological damage of renal tissues was reduced,the concentrations of BUN,Cr,CysC,TNF-αand IL-1βin serum decreased,the level of MDA decreased,the levels of SOD and GSH increased,the mRNA expression levels of
