两种免疫性卵巢早衰大鼠模型的构建与比较

摘  要:目的比较顺铂和雷公藤多苷两种药物制作的免疫性卵巢早衰大鼠模型。方法顺铂模型组按2 mg/kg大鼠体重连续腹腔注射7 d,雷公藤模型组按0.75 mg/kg大鼠体重连续灌胃14 d,补佳乐按0.15 mg/kg连续灌胃21 d进行治疗,观察各组大鼠一般情况、动情周期、性激素水平、卵巢脏器指数、卵巢HE染色和免疫组化。结果末次体重顺铂模型组和顺铂模型治疗组体重低于正常组(P<0.05,n=8);顺铂模型组和雷公藤多苷模型组大鼠动情周期紊乱,顺铂模型治疗组大鼠动情周期为5~8 d/次;顺铂模型组和雷公藤多苷模型组血清促卵泡生成素(FSH)、促黄体生成素(LH)高于正常组,雌激素(E2)低于正常组(P<0.05,n=8);顺铂模型治疗组LH低于顺铂模型组,E2高于顺铂模型组但仍低于正常组(P<0.05,n=8);雷公藤多苷模型治疗组FSH、LH低于雷公藤多苷模型组,E2高于雷公藤多苷模型组(P<0.05,n=8);顺铂模型组、顺铂模型治疗组、雷公藤多苷模型组和雷公藤多苷模型治疗组卵巢脏器指数均低于正常组(P<0.05,n=8);顺铂模型治疗组卵巢指数高于顺铂模型组,雷公藤多苷模型治疗组卵巢指数高于雷公藤多苷模型组(P<0.05,n=8);顺铂模型组和雷公藤多苷模型组卵巢组织HE染色出现闭锁卵泡增多,未见成熟卵泡,顺铂模型治疗组可见卵泡数增多,未见成熟卵泡,雷公藤多苷模型治疗组可见卵泡数增多和成熟卵泡;免疫组化显示顺铂模型组、雷公藤多苷模型组的磷脂酰肌醇3-激酶(PI3K)、蛋白激酶B(AKT)和雷帕霉素的哺乳动物靶标(mTOR)的阳性细胞计数高于正常组(P<0.05,n=8);顺铂模型治疗组mTOR阳性细胞计数低于顺铂模型组(P<0.05,n=8),雷公藤多苷模型治疗组PI3K、pAKT和mTOR的阳性细胞计数低于雷公藤多苷模型组(P<0.05,n=8)。结论两种药物均可制作免疫性卵巢早衰大鼠模型,本次试验雷公藤多苷制作的模型更适合用于药物筛选。Objective To compare the immune premature ovarian failure rat models made by cisplatin and Tripterygium glycosides.Methods The cisplatin groups were injected intraperitoneally at the dose of 2 mg/kg according the weight of rat for 7 days.The model of Tripterygium glycosides groups were orally gavaged at the dose of 0.75 mg/kg according the weight of rat for 14 days.Estradiol was administered orally at the dose of 0.15 mg/kg for 21 days.Both groups were observed the general condition,estrous cycle,sex hormone level,ovarian organ index,ovarian HE staining and immunohistochemical staining.Results The last weight of the cisplatin group and the cisplatin treatment group were lower than that of the normal group(P<0.05,n=8).The rat's estrous cycle in cisplatin group and Tripterygium glycoside group were disordered,while the estrous cycle in cisplatin treatment group and Tripterygium glycoside treatment group was 5~8 days/time.Serum FSH and LH in cisplatin group and Tripterygium glycoside group were higher than those in the normal group,and E2 was lower than those in the normal group,with statistical significance(P<0.05,n=8).LH in the cisplatin group was lower than that in cisplatin group,E2 was higher than that in the cisplatin group,but still lower than that in the normal group,with statistical significance(P<0.05,n=8).FSH and LH in Tripterygium glycoside treatment group were lower than those in Tripterygium glycoside group,and E2 was higher than those in Tripterygium glycoside group,with statistical significance(P<0.05,n=8).The ovarian organ indexes of cisplatin group,cisplatin treatment group,Tripterygium glycoside group and Tripterygium glycoside treatment group were lower than those of the normal group,with statistical significance(P<0.05,n=8).The ovarian index of cisplatin group was higher than that of the cisplatin group,and that of Tripterygium glycoside group was higher than that of Tripterygium glycoside group,with statistical significance(P<0.05,n=8).In cisplatin group and Tripterygium glycoside group,the num
