植物SUS基因家族的系统进化及其在玉米中的干旱诱导表达分析

摘  要:利用生物信息学方法从50个物种内鉴定出283个蔗糖合酶(SUS)家族成员,对鉴定出的SUS家族成员进行系统发育分析、共线性分析、蛋白理化性质和亚细胞定位分析,以及干旱胁迫下玉米SUS基因在不同组织的表达模式分析,综合全面地分析了SUS基因家族的进化规律。系统发育树分析表明:SUS家族可划分为SUSⅠ、SUSⅡ、SUSⅢ三个亚家族,SUSⅢ内部双子叶植物进化枝明显分为两个拓扑结构相似的进化枝;亚细胞定位结果显示SUS蛋白主要在细胞质和细胞核中发挥作用,且SUSⅢ成员的理化性质与SUSⅠ、Ⅱ之间差异明显;共线性结果表明:SUSⅠ、Ⅱ中的共线性基因分别聚成一个独立的亚群,SUSⅢ的共线性基因聚成两个亚群;RNA-seq数据表明:玉米SUS基因Zm00001d045042、Zm00001d029087、Zm00001d029091、Zm00001d014876在干旱胁迫下呈现差异表达,该表达模式分别与各亚家族的基因结构模式较为一致。综合上述研究,推测出植物SUS基因的进化轨迹,即在蕨类植物到种子植物进化过程中分化出SUSⅢ,在被子植物分化以前出现SUSⅠ和SUSⅡ,且SUSⅢ在双子叶植物中具有继续分化的趋势。This project identified 283 Sucrose synthase(SUS)family genes in 50 species.Comphensive analyses of evolutionary relationship of SUS family using bioinformatics methods of phylogenetic analysis,synteny analysis,physicochemical properties analysis,subcellular localization prediction,and expression pattern of maize SUS genes under drought stress were performed.Based on the phylogenetic tree,SUS gene family was divided into three subfamilies,that is SUSⅠ,SUSⅡ,and SUSⅢ.The dicot members of SUSⅢwere clearly divided into two clades with similar topological structures.SUS protein mainly played a role in the cytoplasm and nucleus,and the physical and chemical properties of SUSⅢmembers were different from those of SUSⅠandⅡ.The syntenic genes from SUSⅠand SUSⅡsubfamilies were clustered into one independent community,respectively,and the syntenic genes from SUSⅢsubfamily were clustered into two subgroups.RNA-seq dataset indicated that maize SUS genes Zm00001d045042,Zm00001d029087,Zm00001d029091 and Zm00001d014876 were differentially expressed under drought stress.In addition,the expression pattern of the genes in each subfamily corresponded with their gene structures.Based on the study,the evolutionary trajectory of SUS genes in plants can be inferred that SUSⅢwould differentiate during the evolution of ferns to seed plants,SUSⅠand SUSⅡwould appear before the differentiation of angiosperms,and SUSⅢwould have a tendency to continue to differentiate in dicots.
