摘  要:目的采用病毒显微注射、光遗传、麻醉行为学以及脑电记录等技术从神经环路的角度出发,阐明APC(前梨状皮层,anterior piriform cortex)-IL(侧前额叶下边缘皮层,infralimbic cortex)谷氨酸能通路对全身麻醉的调节作用,并初步探讨其作用机制。方法本研究使用健康雄性C57小鼠,其中6只使用1.4%异氟烷混合氧气(1.5 L/min)麻醉,构建顺行性示踪模型,建模成功后寻找其投射点位。健康C57小鼠16只,在APC处注射光遗传病毒并在IL处埋置光纤,安置脑电电极,建立光遗传学模型,建模成功后记录异氟烷与丙泊酚麻醉的行为学与脑电特征。结果APC处的谷氨酸神经元可以投射到IL处;激活APC^(Glu)-IL通路可延长异氟烷麻醉的诱导时间,减少麻醉特征脑电delta波的比例;但对于丙泊酚麻醉时间与脑电无影响。结论APC^(Glu)-IL通路可以延长异氟烷麻醉的诱导时间,但对丙泊酚麻醉时间无影响。Objective In this experiment,virus microinjection,optogenetic technology,behavioral analysis of anesthesia and EEG recording were used to reveal the functionand possible mechanism of APC-IL glutamatergic pathway in general anesthesia induced consciousness change.Methods Six healthy male C57 mice were anesthetized with 1.4%isoflurane mixed with oxygen(1.5 L/min)to construct an anterograde tracer model to observe the projection sites of APC^(Glu) neurons.Sixteen healthy C57 mice were used to inject rAAV-ChR2 virus in APC and plant the optical fiber in IL and place the EEG electrode on the skull to establish the optogenetics models.After successful modeling,behavior tests of anesthesia and EEG recording were carried out.Results APC^(Glu) neurons could project to IL(infralimbic cortex).In addition,optogenetic activation of APC^(Glu)-IL pathway prolonged the induction time of isoflurane anesthesia and reduced the percentage of delta(δ)wave during isoflurane anesthesia induction,but had no effect on propofol anesthesia.Conclusion APC^(Glu)-IL pathway delays the induction of isoflurane anesthesia,but has no effect on propofol anesthesia.
