摘 要:目的基于UHPLC-Q-Exactive-Orbitrap-MS技术对乌鳖返春口服液的化学成分进行定性分析。方法对纯乙醇沉淀后的乌鳖返春口服液上清液,采用Waters ACQUITY UPLC HSS T3色谱柱(2.1 mm×100 mm,1.8μm),以0.1%甲酸水溶液-乙腈为流动相,梯度洗脱,流速0.30 mL·min^(-1),柱温35℃,进样量4μL,采用HESI源在正、负离子监测模式下采集数据。根据Xcalibur软件的精确相对分子质量、准分子离子峰、碎片离子,结合对照品比对,相关文献和数据库对乌鳖返春口服液中的化学成分进行识别和鉴定。结果乌鳖返春口服液的醇提液中鉴定出131个化学成分,包括三萜类、亚精胺类、萜类、酚酸类、蒽醌类、二苯乙烯类、氨基酸类、黄酮类、脂肪酸类、有机酸类、生物碱类等各类化合物。结论首次采用UHPLCQ-Exactive-Orbitrap-MS对乌鳖返春口服液的化学成分进行了研究,该研究对为乌鳖返春口服液中药效物质基础及作用机制研究提供了重要的理论依据。Objective To qualitatively analyze the chemical compositions of empirical prescription Wubie Fanchun oral liquid by UHPLC-Q-Exactive-Orbitrap-MS.Methods The supernatant of Wubie Fanchun oral liquid was precipitated by ethanol.The analysis was performed on a Waters ACQUITY UPLC HSS T3 column(2.1 mm×100 mm,1.8μm).The mobile phase consisted of 0.1%formic acid water solution and acetonitrile at a flow rate of 0.30 mL·min^(-1.)The column temperature was 35℃and the injection volume was 4μL.The mass spectrometry was acquired with a HESI ion source in both positive and negative ion modes.The components were identified according to the accurate relative molecular weight,excimer ion peak,fragmentation in Xcalibur software,the MS data of reference substances and other information with literature and database.Results Totally 131 components were rapidly identified,including triterpenoids,spermidine derivatives,terpenes,phenolic acids,anthraquinones,distyrene glycosides,amino acids,flavonoids,fatty acids,organic acids,and alkaloids,etc.Conclusion The chemical components of Wubie Fanchun oral liquid have been identified by UHPLC-Q-Exactive-Orbitrap-MS for the first time,which provides the theoretical basis for the study of pharmacodynamic materials basis and mechanism of Wubie Fanchun oral liquid.
