摘  要:为了解蓝莓C_(2)H_(2)锌指蛋白在花芽休眠中的功能,通过生物信息鉴定蓝莓C_(2)H_(2)-ZFP基因家族成员,利用转录组数据和荧光定量PCR分析其在花芽内休眠解除过程和外源脱落酸处理下的表达模式。在蓝莓中共鉴定到VcC_(2)H_(2)-ZFP家族79个成员,通过系统进化树和保守基序将其分为3组。分析表明:VcC_(2)H_(2)-ZFP家族成员在蓝莓各器官中均有表达,其中15个在花芽内休眠解除过程中表达存在显著差异,如Vc31g303.5、Vc7g51.6、Vc16g355.5在深度内休眠时期表达量高,而休眠解除后表达量较低。VcC_(2)H_(2)-ZFP启动子含有大量光反应、防御应激、脱落酸、赤霉素等激素响应元件。外源脱落酸处理花芽能显著促进Vc16g355.5、Vc7g51.6、Vc31g303.5等的表达。本研究结果揭示蓝莓部分VcC_(2)H_(2)-ZFP可能通过响应脱落酸参与花芽休眠过程的调控。In order to explore the function of C_(2)H_(2)zinc finger protein in blueberry flower bud dormancy,members of VcC_(2)H_(2)-ZFP gene family were identified in blueberry by bioinformatics analysis.The expression pattern of C_(2)H_(2)genes during the process of flower bud endodormancy release and treatment of exogenous abscisic acid(ABA)were analyzed through transcriptome data and Quantitative Real-time PCR(qRT-PCR).In this study,79 VcC_(2)H_(2)-ZFP family members were identified in blueberry.According to the results of phylogenetic tree and conservative motif analysis,VcC_(2)H_(2)-ZFP family members were divided into three groups.The results of gene expression showed that VcC_(2)H_(2)-ZFP genes family were expressed in all organs of blueberry,among which 15 VcC_(2)H_(2)-ZFP genes were significantly differentially expressed during flower bud endodormancy release;the expression levels of Vc31g303.5,Vc7g51.6,and Vc16g355.5 were high at deep endodormancy stage,but decreased with the process of dormancy release.Cis-acting element analysis showed that the promoter of VcC_(2)H_(2)-ZFP genes contained a large number of hormone response elements such as photoreaction,stress defense,ABA,and gibberellin.The results of qRT-PCR showed that exogenous ABA treatment of blueberry flower buds can significantly promote the expression of VcC_(2)H_(2)-ZFP genes such as Vc16g355.5,Vc7g51.6,and Vc31g303.5.The results revealed that part of VcC_(2)H_(2)-ZFP might participate in the regulation of blueberry flower bud dormancy by responding to ABA.
