三仙胶囊药物血清对人肾癌786-0细胞株增殖及凋亡的影响

摘　　要：目的通过体外实验观察三仙胶囊药物血清对肾癌细胞生长转移的抑制作用及可能的抑制途径,为体内及临床试验提供理论依据。方法选择240只清洁级Wistar大鼠,按随机数字表分为干扰素组、三仙胶囊组、干扰素三仙胶囊联合组,每组各80只。按血清药理学实验方法获取含药血清。采用三因素3×4×5析因设计及MTT法检测不同作用时间(24、48、72、96、120 h)及不同浓度(5%、10%、15%、20%)的三仙胶囊药物对人肾癌786-0细胞株的生长增殖影响。结果在5%的药物浓度中,三仙胶囊组在96 h时表现出了最高的人肾癌786-0细胞株增殖抑制率,干扰素组与三仙胶囊组的抑制率均在72 h处降至最低,而干扰素三仙胶囊组在48 h后持续上升至96 h,三组抑制率为0.05%~0.35%;在10%的药物浓度中,干扰素组最高抑制率出现在24 h,三仙胶囊组为48 h,而干扰素三仙胶囊组则是96 h,三组细胞抑制率为0.02%~0.18%;在15%的药物浓度中,三仙胶囊组与干扰素三仙胶囊组在48 h处出现最大抑制率,干扰素组则为96 h,三组在72 h处均表现出较低抑制率,抑制率为0%~0.25%;在20%的药物浓度中,三组细胞株增殖抑制率自48 h起呈现出总体下降趋势,干扰素三仙胶囊组在48 h处出现明显上升,三组细胞抑制率为0.03%~0.25%;在不同用药、不同血清浓度以及不同时间的单因素影响的情况下,人肾癌786-0细胞株的增殖抑制率比较,差异均有统计学意义(均P<0.05),不同用药、不同时间以及不同血清浓度、不同时间的两因素间存在交互效应(均P<0.05),同时,用药、血清浓度、时间等三种因素之间共同存在交互效应(均P<0.05)。主体间效应检验的轮廓图并未表现出明显平行迹象。结论三仙胶囊对人肾癌786-0细胞株的生长有明显的抑制作用,三仙胶囊可阻滞人肾癌786-0细胞于S期,从而抑制细胞的增殖;三仙胶囊对人肾癌786-0细胞株有较强的诱导凋亡作用。Objective To observe the inhibitory effect of Sanxian capsule drug serum on the growth and metastasis of renal cancer cells and possible inhibitory pathways through in vitro experiments,so as to provide theoretical basis for in vivo experiments and clinical trials.Methods A total of 240 Wistar rats of clean grade were randomly divided into interferon group,Sanxian capsule group and interferon Sanxian capsule combination group,with 80 rats in each group.The drug-containing serum was obtained according to the method of serum pharmacology experiment.Three factor 3×4×5 factorial design and MTT assay were used to detect the effect of Sanxian capsule drug serum on the growth and proliferation of human renal cancer 786-0 cell line at different treatment time(24,48,72,96,120 hours)and different concentrations(5%,10%,15%,20%).Results The highest inhibition rate of proliferation of 786-0 cells was observed in the Sanxian capsule group at a concentration of 5%at 96 hours.The inhibition rates of the interferon group and Sanxian capsule group were lowest at 72 hours,while the inhibition rates of the interferon group increased from 48 hours to 96 hours.At 10%drug concentration,the highest inhibition rate of interferon group appeared at 24 hours,Sanxian capsule group was 48 hours,and interferon Sanxian capsule group was 96 hours.In general,the three groups showed a low inhibition rate at 72 hours,the inhibition rate increased at 96 hours,and decreased after 96 hours.The inhibition rates of the three groups were 0.02%-0.18%.When the drug concentration was 15%,the maximum inhibition rate of Sanxian capsule group and interferon Sanxian capsule group was 48 hours,and that of interferon group was 96 h.In general,the three groups all showed low inhibition rate at 72 hours,and the inhibition rate was 0%-0.25%.At the drug concentration of 20%,the inhibition rates of cell proliferation in the three groups showed an overall downward trend from 24 hours,and the inhibition rates of interferon Sanxian capsule group increased significantly
