摘  要:目的 研究刺五加苷E(Eleutheroside E,EE)对去卵巢(OVX)小鼠骨量以及神经肽的影响。方法 选取24只7周龄C57BL6/J雌性小鼠,平均分为假手术组(Sham组)、OVX组、EE低剂量组(EE-L组)和EE高剂量组(EE-H组),EE-L组和EE-H组在OVX基础上分别予EE 20 mg/(kg·d)和40 mg/(kg·d)灌胃,Sham组和OVX组以等体积1%羟甲基纤维素钠(CMC-Na)灌胃。12周后收集小鼠左侧股骨和血清标本。左侧股骨行Micro-CT扫描;ELISA法检测血清中骨吸收指标抗酒石酸酸性磷酸酶(TRAP)、骨形成指标Ⅰ型原胶原氨基端肽(P1NP)、炎性细胞因子肿瘤坏死因子-α(TNF-α)、白细胞介素6(IL-6)、白细胞介素10(IL-10)以及神经肽降钙素基因相关肽(CGRP)、神经肽P物质(SP)、血管活性肠肽(VIP)、神经肽Y (NPY)的含量。结果 与Sham组相比,OVX组小鼠骨密度(bone mineral density, BMD)、骨体积分数(BV/TV)、骨小梁数量(Tb.N)、P1NP、IL-10、CGRP、SP、VIP和NPY显著性降低(P<0.05,P<0.01),骨小梁分离度(Tb.Sp)、TRAP、TNF-α和IL-6显著性增高(P<0.05);与OVX组相比,EE-L组小鼠Tb.N、VIP和NPY显著性增高(P<0.05),BMD、BV/TV、IL-10、CGRP和SP有增加趋势,但无显著性差异(P>0.05),Tb.Sp、TRAP、TNF-α和IL-6有降低趋势,但无显著性差异(P>0.05),P1NP未见改变趋势(P>0.05);EE-H组小鼠BMD、BV/TV、Tb.N、P1NP、IL-10、CGRP、SP、VIP和NPY显著性增高(P<0.05,P<0.01),Tb.Sp、TRAP、TNF-α和IL-6显著性降低(P<0.05)。结论 EE能够显著增加OVX骨量,其机制可能是通过降低炎症反应,增加神经肽的含量而实现,并呈现剂量依赖性。Objective To investigate the effects of eleutheroside E(EE)on bone mass and neuropeptides in ovariectomized(OVX)mice.Methods Twenty-four 7-week-old C57BL6/J female mice were randomly divided into Sham group(Sham),OVX group,EE low-dose group(EE-L),and EE high-dose group(EE-H).On the basis of OVX,mice in EE group received 20 mg/(kg·d)of EE-L and 40 mg/(kg·d)of EE-H by gavage.Mice in Sham and OVX groups were gavaged with an equal volume of 1%carboxymethyl cellulose Na.After 12 weeks,the left femur and serum samples were collected.Micro-CT scan was performed on the left femur.The contents of TRAP,P1NP,TNF-α,IL-6,IL-10,CGRP,SP,VIP,and NPY in serum were detected using ELISA.Results Compared with those in Sham,the levels of BMD,BV/TV,Tb.N,P1NP,IL-10,CGRP,SP,VIP,and NPY decreased significantly in OVX group(P<0.05,P<0.01),while Tb.Sp,TRAP,TNF-αand IL-6 increased significantly in OVX group(P<0.05).Compared with those in OVX,the levels of Tb.N,VIP and NPY in EE-L increased significantly(P<0.05).BMD,BV/TV,IL-10,CGRP,and SP showed upward trend,but there was no significant difference(P>0.05).Tb.Sp,TRAP,TNF-α,and IL-6 showed a declined trend,but there was no significant difference(P>0.05).There was no change trend in P1NP(P>0.05).In EE-H group,BMD,BV/TV,TB.N,P1NP,IL-10,CGRP,SP,VIP,and NPY increased significantly(P<0.05,P<0.01),while Tb.Sp,TRAP,TNF-α,and IL-6 decreased significantly(P<0.05).Conclusion EE significantly increases bone mass after OVX,possibly by reducing inflammation and increasing the content of neuropeptides in a dose-dependent manner.
