摘  要:目的观察项痹通颗粒对颈后肌炎症损伤大鼠的炎症相关因子及过氧化物酶增殖激活受体γ/核转录因子KappaB(peroxisome proliferators-activated receptor-γ/NF-KappaB,PPAR-γ/NF-Κb)信号通路相关蛋白的影响,探讨其可能的作用机制。方法将大鼠随机分为假手术组、模型组、塞来昔布组[0.18 g/(kg·d)]、项痹通颗粒低、中、高剂量组[4.5 g/(kg·d)、9.0 g/(kg·d)、18.0 g/(kg·d)],每组各10只。以手术方式切开术口,用弯头止血钳钳夹C 2~6棘突旁开0.2 cm的颈后肌中段(持续钳夹10秒、后松开间隔10秒、扣满3扣),以建立颈后肌炎症损伤大鼠模型。苏木精—伊红(hematoxylin-eosin,HE)染色观察颈后肌组织的形态学变化;酶联免疫吸附法(enzyme-linked immunosorbent assay,ELISA)检测造模前、造模后第2天(干预前)及干预7天后的肿瘤坏死因子-α(tumor necrosis factor-α,TNF-α)、白细胞介素-1β(interleukin-1β,IL-1β)、前列腺素E 2(prostaglandin E 2,PGE 2)水平;蛋白免疫印迹法(Western blot,WB)检测PPAR-γ/NF-κB的活性成分p65(NF-κB p65)、髓样分化因子88(myeloid differentiation factor 88,MyD88)蛋白表达;斜板试验评价大鼠颈部运动功能。结果与造模前比较,造模后第2天(干预前)TNF-α、IL-1β、PGE 2均显著升高(P<0.05)。干预7天后,与假手术组比较,模型组肌细胞核减少、结构紊乱、变形水肿、大小不一,TNF-α、IL-1β、PGE 2、NF-κB p65、MyD88均显著升高(P<0.05),斜板试验功能角度减小(P<0.05);与模型组比较,项痹通颗粒低、中、高剂量组肌细胞核稍增多、结构变清晰、水肿程度减轻、形状较规则,TNF-α、IL-1β、PGE 2、NF-κB p65、MyD88降低(P<0.05),PPAR-γ升高(P<0.05),斜板试验功能角度增大(P<0.05);与塞来昔布组比较,项痹通颗粒中、高剂量组TNF-α、IL-1β降低(P<0.05),项痹通颗粒低、中、高剂量组PPAR-γ升高(P<0.05),NF-κB p65、MyD88降低(P<0.05)。结论项痹通颗粒可改善大鼠颈后肌炎症�Objective To observe effects of Xiangbitong granule(XBTG)on inflammatory factors and PPAR-γ/NF-κB proteins in rats with inflammatory injury of posterior cervical muscle and explore possible mechanism.Methods Rats were randomly divided into the sham operation group(SOG),the model group(MG),the Celecoxib group(CG),the XBTG low-dose group,the middle-dose group and the high-dose group.The rat model was established by surgical incision and clamping middle segment of posterior cervical muscle by C 2~6 spinous process with hemostatic forceps(continuously clamping for 10 s,loosening interval for 10 s and full buckle for 3 times).The method of HE was used to observe morphological changes of posterior cervical muscle.The method of ELISA was used to detect the levels of TNF-α,IL-1βand PGE 2 before modeling,on the second day after modeling(before intervention)and after 7 days of treatment.The method of WB was used to tested PPAR-γ,NF-κB p65 and MyD88,and cervical motor function was evaluated by inclined plate test.Results On the second day after modeling(before intervention)compared with modeling,TNF-α,IL-1βand PGE 2 were increased significantly(P<0.05)in MG.After 7 days of treatment,myocyte nuclei was decreased,structure was disordered,deformation and edema appeared,size was different,TNF-α,IL-1β,PGE 2,NF-κB p65 and MyD88 were increased and function angle of inclined plate test was decreased substantially(P<0.05)in MG opposite to SOG.In contrast to MG,myocyte nuclei slightly increased,structure was clear,degree of edema was reduced,shape was regular,TNF-α,IL-1β,PGE 2,NF-κB p65 and MyD88 were decreased distinctly(P<0.05)and PPAR-γand function angle was ascended significantly(P<0.05)in the XBTG group.Compare with the CG group,the levels of TNF-αand IL-1βwere descended(P<0.05)in the XBTG middle-dose and the high-dose group,and the levels of NF-κB p65 and MyD88 were descended(P<0.05)and PPAR-γwas went up(P<0.05)in the XBTG group.Conclusion XBTG can improve inflammatory injury of posterior cervical muscle in r
