摘　　要：目的探究巴戟天寡糖对绝经后骨质疏松模型大鼠血清骨转化指标和骨组织结构的影响及可能机制。方法50只雌性SD大鼠随机分为假手术组、模型组、阳性药物对照组(阿仑膦酸钠,7 mg/kg)、巴戟天寡糖低剂量组(25 mg/kg)、巴戟天寡糖高剂量组(75 mg/kg),每组10只,除假手术组外,其余各组大鼠构建绝经后骨质疏松(postmenopausal osteoporosis,PMOP)大鼠模型并给予不同药物干预处理后,测定各组大鼠中血清骨碱性磷酸酶(bone alkaline phosphatase,BALP)、I型前胶原氨基端延长肽(procollagen type 1 N-terminal propeptide,P1NP)、抗酒石酸酸性磷酸酶-5b(tartrate resistant acid phosphatase-5b,TRACP-5b)、Ⅰ型胶原C-末端肽交联(C-terminal telopeptide of type 1 collagen,CTX-1)水平,通过双能X线骨密度分析仪对大鼠腰椎骨密度进行检测,采用微计算机断层扫描技术对股骨远端骨微结构进行分析,三点弯曲法测定骨生物力学参数及制片测量骨组织形态计量学参数,使用蛋白免疫印迹法测定左侧股骨骨组织骨保护素(osteoprotegerin,OPG)、核因子κB受体活化因子配体(receptor activator of nuclear faactor-κB Ligand,RANKL)蛋白相对表达水平;qPCR法检测左侧股骨OPG、RANKL的mRNA表达水平。结果与假手术组相比,模型组血清BALP、P1NP水平、腰椎骨骨密度、股骨相对骨体积、骨小梁厚度、骨小梁数量、荧光标记周长百分率、骨形成率与骨体积之比、矿化沉积率、OPG的蛋白及mRNA表达水平显著降低(P<0.05),TRACP-5b、CTX-1、股骨骨小梁间距、结构模型指数、破骨细胞数目、RANKL蛋白、mRNA水平显著升高(P<0.05);与模型组相比,巴戟天寡糖低、高剂量组及阿仑膦酸钠组大鼠BALP、P1NP水平显著上升(P<0.05),股骨相对骨体积、骨小梁厚度、骨小梁数量、荧光标记周长百分率、骨形成率与骨体积之比、矿化沉积率、OPG的蛋白及mRNA表达水平明显增加(P<0.05),TRACP-5b、CTX-1股骨�Objective To explore the effect and possible mechanism of Morinda Officinalis Oligosaccharide(MO)on serum bone turnover indexes and bone tissue structure in postmenopausal osteoporosis model rats.Methods Fifty female SD rats were randomly divided into 10 rats in 5 groups,which is sham operation group(Sham),model group,positive drug control group(alendronate sodium,7 mg/kg),and Morinda officinalis oligosaccharide low-dose group(LMO,25 mg/kg),Morinda Officinalis Oligosaccharide high-dose group(HMO,75mg/kg).Except for the sham-operated group,the other groups of rats were established postmenopausal osteoporosis(PMOP)rat model and given different drug interventions.Serum Bone alkaline phosphatase(BALP),Procollagen type 1 N-terminal propeptide(P1NP),Tartrate resistant acid phosphatase-5b(TRACP-5b)and C-terminal telopeptide of type 1 collagen(CTX-1)levels were measured.The bone mineral density(BMD)of lumbar vertebrae was detected by dual-energy X-ray absorptiometry,the micro-structure of the distal femur was analyzed by micro-CT,the biomechanical parameters of bone were measured by three-point bending method,and the bone histomorphometry was measured by slices.Parameters,the relative expression levels of osteoprotegerin(OPG)and receptor activator of nuclear factor-κB ligand(RANKL)protein in the left femur were determined by Western Blot;the mRNA expression levels of OPG and RANKL in the left femur were detected by qPCR method.Results Compared with the Sham group,the level of BALP and P1NP,bone mineral density(BMD),bone volume/total volume(BV/TV),trabecular thickness(Tb.Th),trabecular number(Tb.N),percentage of fluorescent label perimeter,bone formation rate to bone volume(BFR/BV),mineral apposition rate(MAR),femoral bone tissue OPG protein and mRNA levels in the model group were significantly decreased,TRACP-5b,CTX-1,femoral trabecular spacing(Tb.Sp),Structural model index(SMI),number of osteoclasts,receptor activator of nuclear factor-κB ligand(RANKL)protein,mRNA levels were significantly increased(P<0.05);compared w
