摘  要:为了研究长链非编码RNA生长抑制特异性转录本5(lncRNA GAS5)在小鼠子宫中的表达及其在子宫内膜蜕膜化中的作用,本试验收集妊娠第1~8天的小鼠子宫组织样本,分离妊娠第4天的小鼠子宫内膜基质细胞(ESC)并通过类固醇构建体外诱导蜕膜化模型,采用实时荧光定量PCR(RT-qPCR)检测GAS5、蜕膜化标志因子蜕膜催乳素相关蛋白(Dtprp)、心脏神经脊衍生物表达转录因子2(Hand2)、骨形态发生蛋白2(Bmp2)、肝/肾/骨型碱性磷酸酯酶(Alpl)和同源异形盒基因A10(Hoxa10)的表达水平,采用荧光原位杂交(FISH)技术检测GAS5在ESC中的表达和定位。结果显示,与妊娠第1天相比,在妊娠第4天小鼠子宫组织中GAS5的表达水平显著升高(P<0.05);与妊娠第4天相比,GAS5的表达水平在妊娠第6~8天又随着蜕膜化的进行逐渐下降(P<0.05);与阴性对照(si-NC组)相比,GAS5沉默会引起ESC中蜕膜化相关的基因Dtprp、Hand 2、Bmp 2、Alpl和Hoxa 10表达水平显著增加(P<0.05或P<0.01);GAS5在ESC和蜕膜ESC的细胞质和细胞核中均有表达。结果表明,GAS5参与调节小鼠早期妊娠和蜕膜化进程。This study investigated the expression of long non-coding RNA growth arrest-specific transcript 5(lncRNA GAS5)in the mouse uterus and its role in endometrial decidualization.Uterine tissue samples were collected from mice at gestation days 1 to 8,and mouse endometrial stromal cells(ESC)at day 4 of gestation were isolated and induced by steroids to construct in vitro induced decidualization model.The expression of GAS5,decidualization marker deciduloprolactin associated protein(Dtprp),heart and neural crest derivatives expressed transcript 2(Hand2),bone morphogenetic protein 2(Bmp2),alkaline phosphatase,liver/bone/kidney(Alpl),and homologous heterologous box gene A10(Hoxa10)was assessed by real-time quantitative PCR(RT-qPCR);and the expression and localization of GAS5 in ESC were detected by fluorescence in situ hybridization(FISH).The results showed that compared with the 1st day of gestation,GAS5 expression level in ESC was significantly increased in uterine tissue of mice on the 4th day of gestation(P<0.05).Compared with the 4th day of gestation,the expression level of GAS5 gradually decreased during the 6th to 8th day of gestation with the progression of decidualization(P<0.05).Compared with the negative control(si-NC group),GAS5 silence significantly increased the expression levels of deciduation-related genes Dtprp,Hand 2,Bmp 2,Alpl and Hoxa 10(P<0.05 or P<0.01).GAS5 was expressed in the cytoplasm and nucleus of ESC and decidual ESC.Thus,GAS5 appears to be involved in the regulation of the early pregnancy and deciduating process of mice.
