摘  要:为了明确山麦冬缓解急性肺损伤(ALI)的作用及其机制,本试验使用脂多糖(LPS)诱导小鼠急性肺损伤进行造模,造模前用不同浓度的山麦冬水提物进行干预治疗,通过测定肺组织湿干重比(W/D)评价小鼠肺水肿程度,通过苏木精-伊红(H.E.)染色观察肺组织病理损伤,通过测定氧化应激指标评价肺部氧化损伤情况,通过ELISA检测肺组织中炎症因子的含量,通过免疫组织化学染色技术和Western blot检测ALI小鼠肺损伤相关蛋白的含量。结果显示,LPS造模后,小鼠的肺组织出现严重的损伤,肺组织W/D、氧化应激指标和炎症因子水平均极显著升高(P<0.01);与LPS组比较,山麦冬治疗组小鼠W/D显著降低(P<0.05或P<0.01),肺组织损伤评分显著降低(P<0.05或P<0.01),小鼠血清和肺组织中LDH活力、MPO活力和NO含量均有不同程度降低(P>0.05,P<0.05或P<0.01),小鼠肺组织中炎症因子IL-1β和TNF-α含量和蛋白表达水平均有不同程度降低(P>0.05,P<0.05或P<0.01),小鼠肺组织NF-κB蛋白表达水平显著降低(P<0.05或P<0.01)。结果表明,山麦冬具有良好的防治急性肺损伤的作用,其主要的作用机制可能与激活NF-κB信号通路以减轻炎症反应和抑制氧化应激有关。本试验结果可为山麦冬的ALI缓解作用研究和产业开发提供理论支撑。In order to determine the effect and mechanism of Liriope spicata var.prolifera in alleviating acute lung injury(ALI),this study established a mouse model of ALI with lipopolysaccharide(LPS)induction,utilized water extract of Liriope spicata var.prolifera for intervention treatment,the degree of pulmonary edema was evaluated by measuring the wet-dry weight ratio(W/D)of lung tissue,the pathological injury of lung tissue was observed by hematoxylin-eosin(H.E.)staining,the oxidative stress index was determined to evaluate the oxidative injury of lung tissue,and the content of inflammatory factors in lung tissue was detected by ELISA,the content of lung injury-related proteins in ALI mice was detected by immunohistochemical staining and Western blot.The results showed that the lung tissue of mice had severe injury in ALI animal model induced by LPS,and the levels of lung W/D,oxidative stress indicators and inflammatory factors were significantly increased(P<0.01).Compared with LPS group,W/D in Liriope spicata var.prolifera treatment groups(SMD groups)were significantly decreased(P<0.05 or P<0.01),the scores of lung tissue damage were significantly reduced(P<0.05 or P<0.01),the activity of LDH and MPO and the content of NO in the serum and lung tissue of mice were reduced by varying degrees(P>0.05,P<0.05 or P<0.01),the contents and protein expressions of inflammatory factors IL-1βand TNF-αin the lungs were reduced by varying degrees(P>0.05,P<0.05 or P<0.01);the protein expressions of NF-κB in the lung tissues were significantly reduced(P<0.05 or P<0.01).Thus,L.spicata var.prolifera has a positive effect on acute lung injury,its main mechanism of action may be related to activation of the NF-κB signaling pathway to reduce inflammation and inhibit oxidative stress.This data provides theoretical support for further research and industrial development of L.spicata var.prolifera against acute lung injury.
