摘  要:目的利用低、中、高三个不同的三角波切变率(0 s^(-1)-2.4 s^(-1)-0 s^(-1)、0 s^(-1)-24 s^(-1)-0 s^(-1)和0 s^(-1)-60 s^(-1)-0 s^(-1))的滞后环技术,研究猪肝脱氧核糖核酸蛋白(DNP)悬浮液在三个不同NaCl浓度(在生理盐水基础上加0、1.5、3 mol/L)下的黏弹性与触变性的变化。方法从猪肝制备DNP,采用美国Brookfield公司的程序性DVⅡ流变仪测定DNP样品的滞后环,在Windows下应用该公司的软件,编制自动采样程序,作各种数据(切应力、切变率、温度、时间等)的自动采集、贮存与数据处理。结果在低切变率三角波(0 s^(-1)-2.4 s^(-1)-0 s^(-1))条件下,NaCl浓度提高促使DNP悬浮液具有较高的屈服应力,明显的逆时针滞后环,展示其主要向高黏弹性体方向变化的情景:在中、高切变率三角波(0 s^(-1)-24 s^(-1)-0 s^(-1)和0 s^(-1)-60 s^(-1)-0 s^(-1))条件下,除了上述的性质存在以外,NaCl浓度还促使DNP悬浮液具有较高的触变性,并具有明显的顺时针滞后环。结论NaCl浓度的增高,具有促进DNP水化作用,同时切变率的增高,也是促进DNP水化作用的因素,此两个因素增加了DNP的水化度,致使DNP分子膨胀与改变构型,使之具有较高的黏弹性与触变性。Objective To use the hysteresis loop techniques by the different tri-angular waves with 3 shear rates(0 s^(-1)-2.4 s^(-1)-0 s^(-1),0 s^(-1)-24 s^(-1)-0 s^(-1) and 0 s^(-1)-60 s^(-1)-0 s^(-1)),the behaviors of the visco-elasticity and the thixotropy of deoxyribonucleoprotein(DNP)solution from pig liver were studied under three different concentrations of NaCl with 0,1.5 and 3 mol/L based on the saline water.Methods DNP was prepared from pig liver tissue.The type DV-ⅡProgrammable Rheometer(Brookfield Engineering Laboratories,Inc.,USA)was used for determining datum of hysteresis loops for DNP solution.The experimental datum(including shear rates,shear stress,temperature,time etc)were automatically collected,reserved and treated by the computer program under Windows with the software provided by the Brookfield Engineering Laboratories.Results Under the condition of low shear rate tri-angular waves as 0 s^(-1)-2.4 s^(-1)-0 s^(-1),and under the condition of the increasement of NaCl concentration,the DNP solution possessed high yield stress values and significant anti-clock hysteresis loop areas,which showed that the DNP solution had chiefly turned into higher visco-elastic bodies.And under the condition of middle or high shear rate tri-angular waves as 0 s^(-1)-24 s^(-1)-0 s^(-1),or 0 s^(-1)-60 s^(-1)-0 s^(-1),and also under the condition of the concentration of NaCl,the DNP solution not only possessed high yield stress values and significant anti-clock hysteresis loop areas,which showed that the DNP solution had turned into higher visco-elastic bodies in one hand,but also possessed the along-clock hysteresis loop areas,which showed that the DNP solution had partly turned into higher thixotropic bodies in the other.Conclusion On one hand,to increase the concentration of NaCl in the DNP solution would promote the aquation on the surface of DNP molecules through some hydrophilic groups,and on the other hand,the increase of shear rate also promote the aquation between DNP molecules and water,thus such two factors would
