青海部分县区放牧动物粪便中的ARGs调查及PCR检测

摘  要:随着有机牧场的发展和国家减药行动的开展,相应地区兽用抗生素使用量显著减少,但这是否对该地区放牧动物机体中的抗生素耐药基因(ARGs)产生了影响尚不清楚。为了解青海地区放牧动物体内的ARGs状况,本研究以205份牦牛、藏羊和马的粪便为材料,采用宏基因组测序分析了样本中的ARGs,再从宏基因组分析结果中选择10种ARGs进行了PCR检测。结果显示,祁连县和河南县的牦牛粪便及祁连县的藏羊粪便中检出ARGs 5种;河南、泽库两县的藏羊粪便与海晏县牦牛粪便中检出4种;贵南县牦牛粪便与海晏县藏羊粪便中检出3种;贵南县藏羊中检出2种,在5县区中ARGs检出率最少。结果可能提示被检ARGs在这5县区间存在地区和动物间差异,但需要更多的研究来验证。5种被检出阳性的ARGs中,多重耐药基因OprA在全部粪便样本中的检出率显著高于其它ARGs(P<0.01),可能提示类似OprA的多重ARGs是5县区内的牦牛和藏羊间需要重点关注的耐药基因污染问题,有必要进一步坚持减药行动并采取强化措施。In order to understand the status of ARGs in organic grazing animals in Qinghai Province,the 205 feces samples of yaks,Tibetan sheep and horses were used as materials to analyze the status of ARGs by metagenomic sequencing.Then 10 ARGs were selected from the metagenomic analysis results and detected by PCR.The results showed that 5 species ARGs were detected in yak feces from Qilian County and Henan Mongol Autonomous County and Tibetan sheep feces from Qilian County,4 species were detected in Tibetan sheep dung in two counties of Zeku County and yak dung of Haiyan County.3 species were detected in yak dung in Guinan County and sheep dung in Haiyan County.2 species were detected in sheep dung in Guinan County.The above results may indicate that there were differences of areas and animal among the 5 counties,which suggested that organic ranching construction and national drug reduction action could promote the reduction of ARGs.In addition,among the 5 positive ARGs,the detection rate of the multi-drug resistance gene OprA in all feces samples was significantly higher than that of other ARGs(P<0.01),it is suggested that the multiple ARGs similar to OprA are the major problem of drug resistance gene contamination among yaks and Tibetan sheep in 5 counties,and it is necessary to insist on and further strengthen drug reduction actions and measures.
