摘  要:目的:探讨长托宁对有机磷农药中毒(AOPP)小鼠氧化应激的保护机制。方法:选择健康成年清洁级BALB/c小鼠60只,随机分成敌百虫中毒组(AOPP组,n=10)、对照组(n=10)、预处理组Ⅰ(n=10)、预处理组Ⅱ(n=10)和治疗组Ⅰ(n=10)、治疗组Ⅱ(n=10)。预处理组Ⅰ和Ⅱ先分别肌注0.05 mg·kg^(-1)和0.45 mg·kg^(-1)长托宁,45 min后与AOPP组和治疗组同时使用540 mg·kg^(-1)敌百虫灌胃,出现有机磷农药中毒症状后,治疗组Ⅰ和Ⅱ分别肌注0.05 mg·kg^(-1)和0.45 mg·kg^(-1)长托宁,AOPP组肌注等量生理盐水,对照组进行等量生理盐水灌胃和肌注。观察各组小鼠一般行为情况,眼部取血检测血浆超氧化物歧化酶(SOD)、丙二醛(MDA)水平,观察其胃黏膜显微形态结构变化。结果:对照组一般行为情况良好,AOPP组、治疗组敌百虫灌胃后出现明显中毒症状,治疗组肌注长托宁后症状改善,预处理组灌胃敌百虫后出现轻、中度中毒症状;预处理组Ⅰ、Ⅱ和治疗组Ⅰ、Ⅱ血浆中SOD活性显著高于AOPP组,低于对照组(P<0.05);预处理组Ⅰ、治疗组Ⅰ、Ⅱ血浆中MDA含量显著低于AOPP组,高于对照组(P<0.05);胃黏膜形态结构观察显示,AOPP组胃黏膜严重萎缩变薄,腺细胞萎缩变小、腺腔扩大;预处理组胃黏膜萎缩程度轻于AOPP组,且长托宁浓度越高萎缩程度越轻,治疗组胃黏膜一定程度恢复正常且给药浓度越高恢复越好。结论:长托宁能够通过增强AOPP小鼠SOD活性及细胞膜的氧化耐受性而稳定细胞膜,使MDA含量降低,从而减轻AOPP后机体氧化应激的损伤。Objective:To investigate the protective mechanism of penehyclidine hydrochloride against oxidative stress in mice with acute organophosphate pesticide poisoning(AOPP).Methods:60 healthy adult clean-grade BALB/c mice were randomly divided into trichlorfon poisoning group(AOPP group,n=10),control group(n=10),pretreatment groupⅠ(n=10),pretreatment groupⅡ(n=10),treatment groupⅠ(n=10)and treatment groupⅡ(n=10).The pretreatment groupⅠandⅡwere first injected with 0.05 mg·kg^(-1)and 0.45 mg·kg^(-1)penehyclidine hydrochloride intramuscularly,respectively,and then gavaged with 540 mg·kg^(-1)trichlorfon at the same time as the AOPP group and the treatment group after 45 min.After the symptoms of organophosphate pesticide poisoning appeared,the treatment groupⅠandⅡwere given intramuscular injection of 0.05 mg·kg^(-1)and 0.45 mg·kg^(-1)penehyclidine hydrochloride,respectively,and the AOPP group was given the same amount of normal saline;the control group underwent equal saline gavage and intramuscular injection.All groups of mice were observed for general behavior,and blood was taken from the eyes to detect plasma SOD and MDA levels,and to observe the micromorphological and structural changes of the gastric mucosa.Results:The control group generally behaved well;Obvious symptoms of poisoning occurred after gavage of trichlorfon in the AOPP group and treatment group,and the symptoms improved after intramuscular injection of penehyclidine hydrochloride in the treatment group.Mild and moderate toxicity occurred after gavage of trichlorfon in the pretreatment group;The SOD activity in plasma in the pretreatment groupⅠ,pretreatment groupⅡ,treatment groupⅠ,and treatment groupⅡwas significantly higher than that in the AOPP group and lower than that in the control group,and the difference was statistically significant.MDA levels in the plasma of the pretreat-ment groupⅠ,treatment groupⅠ,and treatment groupⅡwas significantly lower than that of the AOPP group and higher than that of the control group,
