摘 要:可注射生物活性陶瓷不仅具有良好的生物相容性,而且能够促进成骨分化和血管生成。羟基磷灰石(Ca_(10)(PO_(4))_(6)(OH)_(2),HA)是人骨的主要成分。采用微流控技术,成功制备出粒径480μm的均一硅酸钙(CaSiO_(3),CS)微球,并以其为前驱体,在磷酸三钠(Na_(3)PO_(4))水溶液中进行24 h水热转化处理,得到具有纳米线结构的含硅羟基磷灰石微球(Si-nHA)。该纳米线直径约100~200 nm,长度约1~2μm。Si-nHA微球比表面积为7.78 m^(2)/g,远高于纯HA微球的比表面积(0.08 m^(2)/g),高比表面积使得Si-nHA微球具有更加优异的蛋白负载和缓释性能。同时,Si-nHA微球释放的硅(Si)离子能显著促进大鼠骨髓间充质干细胞(rBMSCs)的增殖,以及相关成骨(OPN,OCN)和成血管基因(ANG-1)的表达。研究结果表明,Si-nHA微球在微创骨修复领域有良好的应用前景。Injectable bioactive ceramics not only have good biocompatibility,but also promote osteogenic differentiation and angiogenesis.Hydroxyapatite(Ca_(10)_(PO_(4))_(6)(OH)_(2),HA)is the main component of human bone.Uniform calcium silicate(CaSiO_(3),CS)microspheres with a diameter of about 480μm were successfully prepared using microfluidic technology,and they were used as precursors to prepare silicon-containing hydroxyapatite microspheres(Si-nHA)with nanowire structures in an aqueous solution of trisodium phosphate(Na_(3)PO_(4))by hydrothermal treatment for 24 h.The nanowire had a diameter of about 100~200 nm and a length of about 1~2μm.The specific surface area of Si-nHA microspheres was 7.78 m^(2)/g,which was much higher than that of pure HA microspheres(0.08 m^(2)/g).The high specific surface area resulted in more excellent protein loading and sustained release performance of Si-nHA microspheres.At the same time,Si ions released by Si-nHA could significantly promote the proliferation of rat bone marrow mesenchymal stem cells(rBMSCs)and the expression of related osteogenic(OPN,OCN)and angiogenic genes(ANG-1).The results show that SinHA microspheres have good application prospects in the field of minimally invasive bone repair.
