摘 要:为确定车前草中的总黄酮提取工艺以及总黄酮对腹泻治疗的作用,通过单因素分析实验法和响应面法对车前草总黄酮的提取工艺进行分析和优化。实验采用蓖麻油诱导小鼠腹泻,灌服0.2 mL蓖麻油建立小鼠腹泻模型,进而分析研究车前草中的黄酮类物质在小鼠腹泻中的影响。经研究得出,车前草中的总黄酮提取效果最佳的条件是:(1)乙醇的浓度为70.36%;(2)提取实验过程中温度应控制在85.6℃;(3)原料和提取液的比例应为1∶15.99。在最优工艺条件下每克车前草提取的黄酮的量为12.41 mg·g^(-1),纯度为67.34%。此外,车前草黄酮能显著降低蓖麻油诱导小鼠的腹泻次数、延缓肠道碳粉推进,具有良好的抗腹泻活性。研究为车前草的合理利用以及开发提供了理论基础。In order to determine the extraction process of total flavonoids from plantain and the effect of total flavonoids on diarrhea treatment,the extraction process of total flavonoids from plantain was analyzed and optimized by single factor analysis and response surface methodology.Castor oil was used to induce diarrhea of mice,and 0.2 mL castor oil was infused to establish a mouse diarrhea model,and then the effect of flavonoids in plantain on diarrhea of mice was analyzed and studied.The results showed that the best conditions for the extraction of total flavonoids from plantain were as follows:(1)the concentration of ethanol was 70.36%;(2)during the extraction experiment,the temperature should be controlled at 85.6℃;(3)the ratio of raw material to extract should be 1∶15.99.Under the optimum conditions,the amount of flavonoids extracted from plantain per gram was 12.41 mg·g^(-1) and the purity was 67.34%.In addition,plantain flavonoids could significantly reduce the number of diarrhea induced by castor oil,delay the advancement of intestinal toner with good anti-diarrheal activity,which would provide a theoretical basis for the rational utilization and development of plantain.
