摘 要:为比较猪制品中非洲猪瘟病毒(ASFV)核酸提取效率,将非洲猪瘟假病毒掺入新鲜猪肉、香肠、血粉试验材料,制备相关模拟感染样品,然后采用核酸免抽提、柱式核酸提取及磁珠吸附核酸提取方法提取ASFV核酸,计算ASFV核酸回收率,评估提取效率。结果显示:猪制品模拟感染样品中,ASFV核酸在上清和沉淀中均被检出,且上清中的检出量高于沉淀;对于香肠模拟感染样品上清,3种核酸提取方法均适合,但以柱式提取方法为佳,病毒回收率高达82.8%;对于新鲜猪肉模拟感染样品上清,以磁珠吸附核酸提取方法为佳,病毒回收率为20.9%;对于血粉模拟感染样品上清,以柱式核酸提取方法为佳,病毒回收率为10.2%。本研究为不同猪制品中ASFV核酸提取提供了参考。In order to evaluate the efficiency for extracting nucleic acids of African swine fever virus(ASFV)from pig products,the pseudoviral particles were mixed with the experimental materials including fresh pork,sausage and blood powder to prepare relevant simulated infection samples,and then ASFV nucleic acids were extracted by nucleic acid free extraction,column nucleic acid extraction and magnetic bead adsorption nucleic acid extraction methods,to calculate the recovery rate and to evaluate the extraction efficiency.The results showed that ASFV nucleic acids could be detected from the supernatant and precipitation of the simulated samples,and more in the former as compared with the latter.All the three methods were suitable for the supernatant of sausage samples,in which the column extraction method was the best,with a virus recovery rate of 82.8%;magnetic bead adsorption nucleic acid extraction method was the best for the supernatant of fresh pork samples,with a recovery rate of 20.9%;and the column nucleic acid extraction method was for that of the blood powder samples,with a recovery rate of 10.2%.A reference was thus provided for extracting ASFV nucleic acids from different pig products.
