摘  要:目的:从成体小鼠肝脏中分离培养肝干细胞,并分析其分化潜能。方法:利用门静脉灌流消化法从成体小鼠肝脏中分离细胞并进行体外长期培养,通过脾结节形成实验对其增殖和分化潜能作进一步分析。结果:从成体小鼠肝脏中分离的细胞呈集落样生长,不仅可稳定传代,而且移植至经致死剂量照射的小鼠体内能够形成脾结节,结节中含有肝细胞和胆管上皮细胞特异性标志的细胞。结论:成体小鼠肝脏中存在具有多潜能性的肝干细胞,该细胞的体外成功培养为肝干细胞生物学特性的研究及肝干细胞的应用奠定了基础。Objective: To separate hepatic stem cells (HSCs) from adult mouse liver and to study their differentiation potentiality. Methods: Cells isolated from portal vein of adult mouse liver by purfusion digestion were long-term cultured in vitro. Their characteristics were identified by forming of splenic hyperplasia nodules and immunohistochemistry. Results: Cells isolated from adult mouse liver grew as colonies in vitro and could be subcultured stably. After the cells were transplanted intravenously into irradiated syngeneic mice, a number of hyperplasia nodules were seen in their spleens on the 12th day, in which many cells were positively stained with specific marker of hepatocytes and bile duct cells. Conclusion: The results suggest the existence of bipotential hepatic stem cells in adult mouse liver, and their successful subculture paves the way for further study.
