摘 要:目的:探究胰岛素受体底物-1(IRS1)基因单核苷酸多态性(SNP)位点rs7578326与2型糖尿病合并高甘油三酯的关系,揭示不同基因型对2型糖尿病合并高甘油三酯的影响。方法:采用问卷调查、临床研究、Sanger测序等分子生物学研究方法,将IRS1基因rs7578326位点多态性作为目标,采用SPSS统计分析,确定不同组间的基因型和等位基因频率差异。结果:IRS1基因rs7578326位点等位基因为A、G,等位基因A、G在T2DM+高甘油三酯组和对照组的分布频率依次为85.8%、14.2%,76.3%、23.7%,在高甘油三酯组和T2DM组的分布频率依次为83.3%、16.7%,76.1%、23.9%。与对照组相比,T2DM+高甘油三酯组和高甘油三酯组等位基因频率差异显著(P<0.05),T2DM组无显著差异(P>0.05)。T2DM+高甘油三酯组中AG+AA型人群TG、LDL-C水平均显著高于GG型(P<0.05)。结论:IRS1基因rs7578326位点等位基因A与蒙古族2型糖尿病合并高甘油三酯关联。Objective:To explore the relationship between the single nucleotide polymorphism(SNP)of rs7578326 site in insulin receptor substrate 1(IRS1)gene and type 2 diabetes mellitus(T2DM)complicated with high tri⁃glyceride,and to study the influence of different genotypes on T2DM complicated with high triglyceride.Methods:Applying molecular biology research methods such as questionnaire,clinical research and Sanger sequencing,taking the rs7578326 polymorphism of IRS1 gene as target,and SPSS statistical analysis was used determine the differences of genotype and allele frequency among different groups.Results:The allele of rs7578326 in IRS1 gene was A、G.The frequencies of allele A and G in T2DM+high triglyceride group and control group were 85.8%,14.2%,76.3%and 23.7%respectively.The distribution frequencies in high triglyceride group and T2DM group were 83.3%,16.7%,76.1%and 23.9%respectively.Compared with the control group,the difference of allele frequency between T2DM+high triglyceride group and high triglyceride group was statistically significant(P<0.05),but it was not statistically significant in T2DM group(P>0.05).TG and LDL-C levels in AG+GG group were significantly higher than those in GG group in T2DM+high triglyceride group(P<0.05).Conclusion:Allele A at rs7578326 of IRS1 gene is associated with Mongolian T2DM complicated with high triglyceride.
