固本祛湿化瘀方联合化疗对结肠癌荷瘤小鼠的疗效

摘　　要：目的 观察固本祛湿化瘀方联合奥沙利铂治疗结肠癌荷瘤小鼠的疗效,并初步探讨机制。方法 构建结肠癌荷瘤小鼠模型,随机分为模型组、固本祛湿化瘀方低、中、高剂量共4组。观察各组小鼠体质量、肿瘤体积、肝肺转移及生存情况,并检测外周血T淋巴细胞表达水平。结果 给药期间,中剂量组小鼠体质量较模型组下降缓慢,活动度亦改善;给药结束,各剂量组存活率优于模型组,且低、中剂量组生存时间明显延长,CD4^(+)、CD8^(+)T淋巴细胞表达水平明显升高。结论 固本祛湿化瘀方虽未明显增强奥沙利铂治疗结肠癌荷瘤小鼠的疗效,但可显著改善小鼠的体质量及活动度,延长总生存时间,其作用机制可能与提高小鼠T淋巴细胞水平,改善免疫功能有关。Objective To observe the curative effect of Guben Qushi Huayu recipe assisted by oxaliplatin in the treatment of colon cancer tumor⁃bearing mice,and to preliminarily explore its mechanism.Methods A colon cancer tumor⁃bearing mouse model was constructed.The mice were randomly divided into model group,low⁃dose,medium⁃dose and high⁃dose groups of Guben Qushi Huayu recipe.The effect of different dose groups on the body weight,tumor volume the lung and liver metastasis and survival rate was observed.The expression level of T lymphocytes in the peripheral blood of the mice in each group was detected.Results During the administration period,the body weight of the mice in the middle⁃dose group decreased significantly compared with the model group,and the activity of the mice was also significantly improved compared with the model group.After administration,the survival rate of mice in the each dose group of Guben Qushi Huayu recipe was significantly higher than that of the model group.Compared with the model group,the survival time of the mice in the low and medium dose groups of traditional Chinese medicine was significantly prolonged,and the expression levels of CD4^(+)T lymphocytes and CD8^(+)T lymphocytes were significantly increased.Conclusion Guben Qushi Huayu recipe does not significantly enhance the efficacy of oxaliplatin in the treatment of colon cancer tumor⁃bearing mice,but can significantly improve the body weight and activity of tumor⁃bearing mice,and prolong the overall survival time.Its mechanism of action may be related to increasing the level of T lymphocytes in mice and improving the immune function of mice.
