摘 要:WRKY转录因子家族参与高等植物生理过程,并在保护植物免受非生物胁迫方面起到至关重要的作用。本研究基于小兰屿蝴蝶兰(Phalaenopsis equestris)基因组数据库对小兰屿蝴蝶兰进行WRKY基因家族成员鉴定、进化关系、保守motif分析、基因结构、功能启动子元件和基因表达分析。结果表明,共鉴定出57个WRKY家族基因,分为GroupⅠ、GroupⅡ和GroupⅢ,分别有12、37和8个成员,GroupⅡ可进一步分为Ⅱa、Ⅱb、Ⅱc、Ⅱd、Ⅱe亚组。小兰屿蝴蝶兰WRKY基因家族成员氨基酸数量长度为76~565 aa,平均长度为270 aa,等电点为4.12~10.75。保守motif分析表明,各组成员的保守基序基本一致,但有6个PeWRKY的WRKYGQK结构域发生了变异,占小兰屿蝴蝶兰WRKY家族基因的10.5%。基因结构分析表明,PeWRKY家族基因包含1~6个内含子和2~7个外显子。功能启动子元件分析表明,在PeWRKY家族基因的启动子区域发现了大量与胁迫响应或植物激素相关的顺式作用元件。基因表达分析表明,55个PeWRKY至少在一个组织中表达。本研究利用生物信息学分析手段,发现与小兰屿蝴蝶兰生长发育和胁迫响应相关的WRKY基因,为进一步研究小兰屿蝴蝶兰的抗逆性和分子育种提供有价值的信息。The WRKY transcription factor family participates in the physiological processes of higher plants and plays a vital role in protecting plants from abiotic stress.In this study,based on the Phalaenopsis equestris genome database,the WRKY gene family member identification,evolutionary relationship,conservative motif analysis,gene structure,functional promoter elements and gene expression analysis of P.equestris were carried out.The results showed that a total of 57 WRKY family genes were identified,divided into GroupⅠ,GroupⅡand GroupⅢ,with 12,37 and 8 members respectively.GroupⅡcould be further divided intoⅡa,Ⅱb,Ⅱc,Ⅱd andⅡe subgroups.The number of amino acids in the WRKY gene family members of P.equestris was 76~565 aa,the average length was 270 aa,and the isoelectric point was 4.12~10.75.Conservative motif analysis showed that the conserved motifs of the members of each group were basically the same,but there were 6 genes of the PeWRKY WRKYGQK were mutated,accounting for 10.5%of the WRKY family genes in P.equestris.Gene structure analysis showed that PeWRKY family gene contained 1 to 6 introns and 2 to 7 exons.The analysis of functional promoter elements showed that a large number of cis-acting elements related to stress response or plant hormones were found in the promoter region of PeWRKY family gene.Gene expression analysis showed that 55 PeWRKY were expressed in at least one tissue.This study used bioinformatics analysis methods to find the WRKY gene that responds to the growth and development and response of the P.equestris,providing valuable information for further research on the stress resistance and molecular breeding of the P.equestris.
