摘 要:风信子作为春季园林景观中的流量花卉,应用方式灵活多样。由原种风信子(Hyacinthus orientalis)培育而来的现代风信子品种资源极为丰富,部分园林常用品种存在同物异名或同名异物现象,本研究利用AFLP分子标记对风信子园艺品种进行有效分类,为新品种选育提供新思路。利用64对引物对30个常见园艺风信子品种进行AFLP分子标记测定,使用NTSYSpc2.1软件计算其遗传相似系数。并通过NTSYSpc2.1软件中的UPGMA法构建聚类树状图。筛选出的8对AFLP引物通过PCR扩增获得条带11703条,其中多态性条带9033条,多态性比率为77.18%;所有品种的遗传相似系数范围在0.7969~0.9167之间,平均值为0.8649。以相似系数0.8300为阈值,30个风信子品种可分为两大类群、两个亚类、五个组群,其中,‘White Pearl’和‘Carnegie’相似系数最大,为0.9167;‘Jan Bos’和‘Atlantic(2008)’相似系数最小,为0.7969,小组群多个品种间的遗传系数均达到0.9000以上,如‘Sky Jacket’与‘Aiolos’(0.9115)、‘Purple Sensation’与‘China Pink’(0.9005)、‘Pink Pearl’与‘Blue Pearl’(0.9051)。聚类结果发现相同色系、倍性的品种几乎聚为一类,异色系中紫色系常作为中间色与其他色系聚为一类。通过AFLP分子标记鉴定的现代风信子园艺品种均具有较高的遗传多样性水平,遗传背景较窄,相同色系和倍性的园艺品种间亲缘关系最近,因此,可选用不同花色和倍性的风信子品种作为亲本进行优良新品种培育。As a frequently-used flower in spring landscape,Hyacinth orientalis is of various application modes.The germplasm resources of the protospecies-Hyacinth orientalis have changed greatly,synonymy is a common phenomenon that occurs among common horticultural hyacinth cultivars.In this study,AFLP molecular markers were used to effectively classify horticultural hyacinth cultivars and provide new ideas for new cultivar breeding.64 pairs of primers of AFLP molecular markers were used to analyze 30 common horticultural hyacinth cultivars,NTSYSpc2.1 was applied to UPGMA clustering analysis to construct the phylogenetic tree.8 pairs of AFLP primers amplified a total of 11703 clear bands,of which 9033 were polymorphic,and the percentage of polymorphic bands was 77.18%.The genetic similarity coefficient among 30 hyacinth cultivars ranged from 0.7969 to0.9167,with the average of 0.8649.By fixing the value of 0.8300 as the threshold of genetic similarity coefficient,30 hyacinth cultivars can be divided into two broad categories,two subclasses and five groups.Among them,'White Pearl'and'Carnegie'had the highest genetic similarity coefficient(0.9167),while Jan Bos and'Atlantic(2008)'had the lowest similarity coefficient(0.7969).The genetic similarity coefficients among some cultivars in the group were all above 0.9000.Examples include'Sky Jacket'and'Aiolos'(0.9115),'Purpose Sensation'and'China Pink'(0.9005),Pink Pearl and'Blue Pearl'(0.9051).The clustering results showed that the cultivars with the homo-chromatic system and ploidy were almost clustered into one group,and the purple system was often clustered into one group with other color systems as the intermediate color system.Modern hyacinth horticultural cultivars identified by AFLP molecular markers all have high level of genetic diversity,theirs genetic background is narrow,and the closest genetic relationship between horticultural cultivars with the same color and ploidy.Therefore,hyacinth cultivars with different color and ploidy can be selected as parents to breed exc
