摘 要:本试验以热敏型生菜‘GRAND RAPIDS TBR’为材料,分别探讨不同消毒试剂及时间对种子消毒的影响及不同生长调节剂对愈伤组织诱导、不定芽再生的影响,初步建立了高效的再生体系。结果表明:种子消毒的最适配比为75%的酒精45 s+3%的次氯酸钠12 min,种子成活率可达97.8%;愈伤组织诱导与不定芽分化的最适培养基为MS+0.1 mg/L 6-BA+0.5 mg/L NAA,愈伤组织诱导率100%,平均不定芽再生27.7个,且不定芽生长健壮;最适生根培养基为不添加NAA的纯MS培养基,生根率为97.8%;炼苗移栽后,植株成活率100%。本研究初步建立了‘GRAND RAPIDS TBR’的高效再生体系,为生菜基因功能研究和耐高温新品种培育提供基础。In order to establish an efficient regeneration system of heat sensitive lettuce,'GRAND RAPIDS TBR'was used as the explant to study the effectives of different sterile reagent disinfectants and disinfection time on the seeds sterilization and the effectives of different combinations of plant growth regulators on callus induction and shoot differentiation.The results showed that the most optimal combination for seed sterilization was 75%alcohol for 45 s+3%NaClO for 12 min,and the survival rate of seeds was 97.8%;the optimal medium for callus induction and adventitious shoots differentiation was MS+0.1 mg/L 6-BA+0.5 mg/L NAA.The callus induction rate was100%.The average number of adventitious shoots was 27.7 and the adventitious shoots were differentiated strongly.The optimal medium for rooting was the MS culture medium without NAA,the rooting rate was about 97.8%;after refining seedlings and transplanting,the survival rate of plants was 100%.In this study,the efficient regeneration system of heat sensitive lettuce'GRAND RAPIDS TBR'was established,which provided a basis for the study of gene function and the cultivation of new varieties resistant to high temperature of lettuce.
