摘 要:本试验以美国红枫带芽茎段为外植体,通过筛选外植体消毒方式,筛选出适宜的初代诱导培养、继代增殖培养、壮苗培养、生根培养的培养基,建立美国红枫组织培养体系,从而为美国红枫工厂化育苗提供技术支持。研究结果表明:最佳的消毒处理方式是先后用75%酒精消毒30 s及10%次氯酸钠消毒10 min,平均存活率为64.15%;适宜的腋芽诱导培养基为改良MS+1.0 mg/L 6-BA+0.2 mg/L IBA。在改良MS培养基中添加0.005 mg/L TDZ+0.1 mg/L 6-BA+0.1 mg/L GA3+0.1 mg/L IBA适用于美国红枫继代增殖培养,增殖系数为6.91;筛选最佳的壮苗培养基为MS+1.0 mg/L ZT+0.3 mg/L IBA;生根培养基为1/2MS+1.0 mg/L GA3+2 mg/L 2,4-D,平均生根数量为5条。In this experiment,the American red maple with bud and stem segments as explants,through the screening of explants disinfection,screen out the appropriate culture medium for primary induction culture,secondary proliferation culture,strong seedling culture,rooting culture,to establish the American red maple tissue culture system.Cultivation system to provide technical support for the industrialized nursery of American red maple.The research results showed that the best disinfection method for explants was 75%alcohol for 30 s+10%sodium hypochlorite for 10 min,and the average survival rate was 64.15%;the appropriate axillary bud induction medium was modified MS+1.0 mg/L 6-BA+0.2 mg/L IBA,modified MS medium was better than WPM medium than MS medium.Add 0.005 mg/L TDZ+0.1 mg/L 6-BA+0.1 mg/L GA_3+0.1 mg/L IBA to the modified MS medium,which is suitable for the secondary proliferation culture of American red maple,with a proliferation coefficient of 6.91;the best selection the strong seedling medium is MS+1.0 mg/L ZT+0.3 mg/L IBA,and the subculture period is 45 days;the rooting medium is 1/2MS+1.0 mg/L GA_3+2 mg/L 2,4-D,the average number of roots is 5.
