摘 要:目前,培养人参形成层分生组织细胞(CMCs,也称作干细胞)的材料来源于贮藏根形成层。但植物内生菌感染给后续培养带来了很大的不确定因素。本文以人参无菌苗为原材料培养CMCs,避免了植物内生菌感染的问题。另外,在无菌苗培育阶段,对根尖CMCs进行诱导培养,同时抑制根的分化,又避免了渗透剂的使用。本文构建制备的无菌苗来源CMCs悬浮后多以单细胞或者2~3个细胞的小细胞团的形式存在,单细胞率高达95%,为建立人参CMCs后续工业化生产提供了研究基础。Currently,the material for cultivating ginseng cambium meristem cells(CMCs,also known as stem cells)comes from the storage root.However,infection of plant endophytes brings a great deal of uncertainty in the subsequent culture.In present paper,aseptic seedlings were used as a raw material to avoid the problem of plant endophyte infection.In addition,during the culture of aseptic seedlings,root tip stem cells were induced and root differentiation was suppressed simultaneously.Thus percolation therapy was avoided.The stem cells from aseptic seedlings mostly existed in the form of single cells or tiny cell clusters comprising 2~3 cells after suspension.The single cell rate was as high as 95%.The present work provides a research basis for the industrial production of ginseng CMCs.
