摘 要:目的:运用网络药理学探讨黄连的主要化学活性成分,分析总结黄连在治疗痤疮疾病方面的作用机制。方法:利用TCMSP检索“黄连”(Coptidis Rhizoma)的活性成分,并筛选出其活性成分所对应的作用靶点。运用GeneCards数据库平台检索与痤疮(Acne)相关作用靶点,构建“药物—疾病”共同靶点Venn图。利用Cytoscape3.9.1软件,绘制“药物活性成分-靶点-疾病”的网络调控图,借助String数据库构建蛋白-蛋白相互作用网络,获得PPI网络核心基因的邻接节点数目,再运用Cytoscape3.9.1对PPI网络进行拓扑分析,最终获取核心基因。通过R语言包cluster Profiler进行GO功能注释和KEGG通路富集分析,预测黄连对痤疮的作用机制。结果:分析预测得到黄连有效成分17个,涉及350个作用靶点。确定痤疮靶点1649个,得到共同靶点基因76个,运用PPI分析确定TNF、IL6、PTGS2、NF-κB/P65、MAPK1、EGF等是治疗痤疮的核心靶点。富集分析GO条目535项(P<0.05),KEGG信号通路233条(P<0.05),主要富集通路有AGE-RAGE信号通路、MAPK信号通路、NF-κB信号通路等。结论:黄连可通过多靶点、多通路治疗痤疮,在发挥抗菌消炎作用的同时还能促进细胞增殖帮助伤口愈合,为黄连在临床的应用提供理论参考和研究方向。Objective:To explore the main active ingredients and mechanism of Coptidis Rhizoma treating acne using network pharmacology.Methods:Using TCMSP to search the active ingredients corresponding in Coptidis Rhizoma,and screen the corresponding targets of the active ingredients.Gene Cards database platform was used to mine the targets related to Acne,and the Venn diagram of the“drug-disease”common target was constructed.Cytoscape 3.9.1 was used to draw the network regulation map of“drug active component-target-disease”,and the protein-protein interaction(PPI)network was constructed with the help of String11.5 to obtain the number of adjacent nodes of core genes in the PPI network.Then Cytoscape 3.9.1 was used for topology analysis of the PPI network,and finally,the core genes were obtained.Functional annotation of gene ontology(GO)and pathway enrichment analysis based on the KEGG was taken usingthe R language package cluster Profiler to predict the mechanism of Acne on promoting fracture healing.Results:17 active components involving 350 targets were predicted in Coptidis Rhizoma.1649 targets for Acne were identified and 76 common target genes were obtained.The PPI analysis was used to identify TNF、IL6、PTGS2、NF-κB/P65,MAPK1,EGF as the core targets for promoting Acne.Enrichment analysis included 535 GO items(P<0.05)and 233 KEGG signaling pathways(P<0.05).The main enrichment pathways were AGE-RAGEsignaling pathway,MAPK signaling pathway,NF-κB signaling pathway,etc.Conclusion:Coptidis Rhizoma can treat acne through multiple targets and pathways.It can not only play an antibacterial and anti-inflammatory role,but also promote cell proliferation to help wound healing,providing theoretical reference and research direction for the clinical application of Coptidis Rhizoma.
