摘 要:目的:探讨清化凉血护络汤对大鼠急性放射性肠炎的预防作用及其可能机制。方法:将36只健康成年SPF级Wistar大鼠随机分为对照组、模型组、低剂量及高剂量组,每组9只。高剂量组按0.08 g·ml^(-1)·kg^(-1),低剂量组按稀释4倍后剂量,提前予清化凉血护络汤灌胃1周,2次·d^(-1),1 ml·次^(-1)。除对照组外,其余各组采用高能X射线进行辐照,总剂量17.5 Gy,建立大鼠急性放射性肠炎模型。照射后12 h高剂量组、低剂量组均继续灌胃给药10 d;对照组和模型组大鼠给予相同体积生理盐水灌胃10 d。每日观察大鼠一般状况和排便情况,各组分别于照射后第4、7、10天取空肠、直肠上段组织及腹主动脉血清,光镜下观察肠壁各层形态学变化并计算单位面积空肠肠腺存活率,采用Hovdenak评分系统对直肠量化评分,并用ELISA法测定环氧合酶-2(COX-2)、肿瘤坏死因子-α(TNF-α)、白细胞介素-6(IL-6)和热休克蛋白90α(Hsp90α)含量。结果:低剂量组及高剂量组一般征象评分、直肠Hovdenak评分明显低于模型组(P<0.01),回肠肠腺存活率明显高于模型组(P<0.01);低剂量组与高剂量组相比,一般征象评分、直肠Hovdenak评分、回肠肠腺存活率差异有统计学意义(P<0.01);建模给药第10天,低剂量组和高剂量绒毛高度、隐窝深度均高于模型组,差异有统计学意义(P<0.05)。高剂量组大鼠血清COX-2、TNF-α、IL-6、Hsp90α含量显著低于模型组(P<0.01)。结论:清化凉血护络汤能有效改善大鼠急性放射性肠炎的症状,减轻及修复肠黏膜损伤,作用机制可能与降低炎症因子、减少细胞凋亡有关。Objective:To investigate the preventive action and possible mechanism of Qinghua Liangxue Huluo decoction in treating rats with acute radiation enteritis.Methods:Thirty-six healthy adult SPF Wistar rats were randomly divided into control group,model group,low-dose group and high-dose group,with 9 rats in each group.The high-dose group was given a dose of 0.08 g·ml^(-1)·kg^(-1),and the low-dose group was given a 4-fold diluted dose.Qinghua Liangxue Huluo decoction was given by intragastric administration for 1 week in advance,2 times·d^(-1),1 ml·time^(-1).Except for the control group,the other groups were given a total dose of 17.5 Gy of high energy X-rays on the abdominal region to establish acute radiation enteritis models.Twelve hours later,the high-dose group and the low-dose group were assigned intragastric administration for ten days.The Rats in the control group and the model group were given the same volume of normal saline for 10 days.The general condition and defecation in all groups rats were observed every day.Samples from the jejunum,upper rectum,and abdominal aortic serum were taken on days 4,7,and 10 to examine pathological changes and calculate the crypt survival rate.The rectum was quantified by Hovdenak scoring system,and the levels of cyclooxygenase-2(COX-2),tumor necrosis factor(TNF-α),interleukin-6(IL-6),and heat shock protein 90α(Hsp90α)were measured by enzyme-linked immunosorbent assay.Results:The scores of general signs and rectal Hovdenak in the low dose group and the high dose group were significantly lower than those in the model group(P<0.01),and the survival rate of ileal gland in the low dose group and the high dose group were significantly higher than that in the model group(P<0.01).The scores of general signs,rectal Hovdenak and survival rate of ileal gland in the low dose group had significant difference with the high dose group(P<0.01).On the 10th day of modeling and administration,the villus height and crypt depth in the low dose group and the high dose group were signific
