摘 要:合成了一种绿色荧光碳量子点(CD),使用分子量为25 kDa的支化聚乙烯亚胺修饰CD表面,通过静电作用得到CD-聚乙烯亚胺(PEI)体系,该体系具有良好的生物相容性和稳定的光学性质。以CD-PEI为载体,实现抗肿瘤基因p53的递送。通过Zeta电位和紫外光谱的变化以及细胞成像图可知,p53质粒成功与CD-PEI体系结合。分别用CD-PEI/p53纳米体系孵育正常细胞和肝癌细胞,通过3-(4,5-二甲基噻唑-2)-2,5-二苯基四氮唑溴盐(MTT)实验可知该体系对肝癌细胞增殖具有较强的抑制效果,而对正常细胞的毒性较小,故可用于肝癌细胞的治疗。同时,与传统PEI/p53组比较,CD-PEI/p53纳米体系具有更高的转染效率和增殖抑制率,其转染效率是传统PEI/p53组的1.6倍,增殖抑制率比传统PEI/p53组的高12%。Green fluorescent carbon dots(CDs)were synthesized.The surface of CDs was modified by branched polyethyleneimine with a molecular weight of 25 kDa.The CDs-polyetyleneimine(PEI)system was obtained by electrostatic interaction.The system has good biocompatibility and stable optical properties.CDs-PEI was used as the vector to deliver the anti-tumor gene p53.According to the changes of Zeta potential and UV spectrum,as well as cell imaging,it can be seen that p53 plasmid successfully combines with CDs-PEI system.The normal cells and liver cancer cells were incubated with CDs-PEI/p53 nano system,respectively.3-(4,5-Dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide(MTT)experiment shows that the system has a strong inhibitory effect on the proliferation of liver cancer cells,but has less toxicity to normal cells,thus it can be used for the treatment of liver cancer cells.Meanwhile,compared with the traditional PEI/p53 group,the CDs-PEI/p53 nano system has higher transfection efficiency and proliferation inhibition rate.The transfection efficiency of the CDs-PEI/p53 nano system is 1.6 times that of the traditional PEI/p53 group,and the proliferation inhibition rate of the nano system is 12%higher than that of the traditional PEI/p53 group.
