摘 要:目的探讨黄芩苷对糖尿病大鼠血管损伤的保护作用及机制。方法40只大鼠随机分为4组,分别为空白组、糖尿病组、黄芩苷50 mg/kg组、黄芩苷100 mg/kg组。单次腹腔注射链脲佐菌素65 mg·kg-1·d-1用于建立糖尿病模型,注射链脲佐菌素后黄芩苷灌胃,空白组和模型组给予等量羧甲基纤维素钠灌胃,每日1次,灌胃28 d。离体血管环实验检测大鼠血管内皮功能,ELISA法检测血清中IL-18,IL-1β,TNF-α,IL-6,丙二醛(MDA)和谷胱甘肽还原酶(GSH-px)含量;HE染色观察各组大鼠主动脉形态;硝酸还原酶法检测各组大鼠血清NO水平;免疫印迹检测各组大鼠主动脉组织中eNOS、NF-κB蛋白表达;二氢乙锭(DHE)染色观察各组大鼠主动脉活性氧(ROS)形成情况。结果经黄芩苷治疗后,糖尿病大鼠血管舒张功能障碍改善,血清中IL-β,IL-18,IL-6和TNF-α含量降低,NO、MDA和GSH-px含量升高,主动脉组织中eNOS蛋白表达增加、NF-κB蛋白表达降低,ROS形成减少。结论黄芩苷可能通过NF-κB/ROS信号通路保护链脲佐菌素诱导的糖尿病大鼠血管损伤。Objective To investigate the protective effect and mechanism of baicalin on vascular injury in diabetic rats.Methods Forty rats were randomly divided into 4 groups:a control group,a diabetes group,a baicalin 50 mg·kg-1 group,and a baicalin 100 mg·kg-1 group.A single intraperitoneal injection of streptozotocin(65 mg·kg-1·d-1)was used to establish a diabetic model.After injection of streptozotocin,baicalin was perfused.The control group and the model group were given the same amount of carboxymethyl cellulose sodium,once a day for 28 days.Vascular endothelial function of rats was detected by in vitro vascular ring assay.ELISA method was used to detect the serum levels of IL-8,IL-1β,TNF-α,IL-6,malondialdehyde(MDA)and glutathione reductase(GSH-px).HE staining was used to observe the morphology of aorta in each group.The level of NO in serum was measured by nitrate reductase method.The expression of eNOS and NF-κB protein in aorta of rats in each group was detected by Western blotting.The formation of reactive oxygen species(ROS)in aorta of rats in each group was observed by DHE staining.Results After baicalin treatment,the vasodilatory dysfunction of diabetic rats was improved,the levels of IL-β,IL-8,IL-6,and TNF-αin the serum of rats were decreased,the levels of NO,MDA and GSH-px were increased,the expression of eNOS protein and NF-κB protein in aorta tissue were increased,and the formation of ROS was decreased.Conclusions Baicalin may protect streptozotocin-induced vascular injury in diabetic rats through the NF-κB/ROS signaling pathway.
