摘  要:目的探讨龟鹿补肾丸对过敏性哮喘模型大鼠气道炎症及Th1/Th2失衡的影响。方法48只SD雄性大鼠随机分为正常对照组、模型组、地塞米松阳性药物组、龟鹿补肾丸低、中、高剂量组。除正常对照组外,其余各组采用卵蛋白(OVA)致敏激发制备大鼠过敏性哮喘模型。各给药组从激发第1天开始给予相应药物进行干预。末次激发10 min记录大鼠引喘潜伏期和哮喘症状评分。末次雾化激发24 h后,检测外周血总白细胞数、分类和肺泡灌洗液(BALF)总白细胞数,ELISA法检测血清IgE水平和BALF中的IL-4、IL-5和IL-13和IFN-γ水平,HE染色和ABPAS染色分别观察肺组织炎症改变和气道酸性黏液分泌变化。qPCR法测定肺组织的T-bet、GATA-3的基因表达水平。结果与模型组比较,龟鹿补肾丸能显著降低过敏性哮喘大鼠症状评分、延长引喘潜伏期(P<0.01或P<0.05),显著降低BALF、外周血的总白细胞数以及外周血的淋巴细胞百分比、嗜酸性粒细胞百分比及中性粒细胞百分比(P<0.01或P<0.05),减少支气管周围炎性细胞浸润,酸性黏液物质阳性面积显著减小(P<0.01),明显降低过敏性哮喘大鼠外周血IgE水平(P<0.01),显著升高BALF中IFN-γ水平,降低IL-4、IL-5和IL-13水平(P<0.01或P<0.05),肺组织的T-bet mRNA表达水平显著升高,而GATA-3 mRNA表达水平显著降低(P<0.01或P<0.05)。结论龟鹿补肾丸能减轻过敏性哮喘大鼠气道炎症,其机制可能与抑制GATA-3 mRNA表达,促进T-bet mRNA表达,调节Th1/Th2失衡以缓解过敏性哮喘Th2型炎症有关。Objective To investigate the effect of Guilubushen pill on airway inflammation and Th1/Th2 imbalance in rats with allergic asthma.Methods 48 Male SD rats were randomly divided into the normal control group,the model group,dexamethasone positive drug group,and low,medium and high doses of Guilubushen pill groups.Except the normal control group,the other groups were sensitized with OVA to establish the rat model of allergic asthma.Each administration group was given corresponding drugs from the first day of stimulation for intervention.Asthmatic latency and asthma symptom score were recorded 10 min after the last stimulation.24 h after the last atomization excitation,the total white blood cell count and classification in peripheral blood and total white blood cell count of the alveolar lavage fluid(BALF)were detected.Serum IgE level and the levels of IL-4,IL-5,IL-13 and IFN-γin BALF were detected by ELISA.HE staining and AB-PAS staining were used to observe the changes of lung tissue inflammation and airway acidic mucus secretion.The expression of T-bet and GATA-3 in lung tissues was determined by qPCR.Results Compared with model group,Guilubushen pill significantly reduced the symptom score,prolonged asthmatic latency of rats(P<0.01 or P<0.05),and reduced the total white blood cell counts in BALF and peripheral blood,and percentages of lymphocytes,eosinophils and neutrophils in peripheral blood(P<0.01 or P<0.05).Guilubushen pill clearly decreased the inflammatory cell infiltration around bronchus,the positive area of acidic mucus substance(P<0.01),and the IgE level in peripheral blood(P<0.01).Meantime,the IFN-γlevel in BALF was significantly increased,and the levels of IL-4,IL-5 and IL-13 in BALF were decreased(P<0.01 or P<0.05).The expression of T-bet mRNA in lung tissues was obviously increased,while the expression of GATA-3 mRNA was lowered(P<0.01 or P<0.05).Conclusion Guilubushen pill can reduce airway inflammation in rats with allergic asthma,which may be related to inhibition of GATA-3 expression,upregulat
