半红树植物黄槿的组织培养

摘　　要：以半红树植物黄槿茎尖为外植体,探讨激素浓度对不定芽的诱导和增殖以及不定根诱导的影响,建立其组织培养体系。结果表明:(1)消毒方法为75%酒精消毒30 s,无菌水冲洗3次,接着用0.1%HgCl_(2)消毒6 min,无菌水冲洗5次;(2)不定芽增殖培养基最佳配比为WPM+0.5 mg·L^(-1)6-BA+0.1 mg·L^(-1)NAA;(3)生根培养基最佳配比为1/2WPM+0.3 mg·L^(-1)IBA+0.3 mg·L^(-1)NAA;(4)驯化完毕的组培苗移植于装有混合基质中(泥炭土∶黄心土=1∶1)的营养钵中,移植成活率为100%。本文相关数据可为黄槿良种选育及遗传转化等研究提供技术支撑。In order to establish a tissue culture system for mangrove associate species H.tiliaceus,the shoot apex was used as explant source to study the effects of phytohormone combination on the proliferation of adventitious buds and the induction of adventitious roots in the present study.The results showed that:(1)the disinfection protocol was 75% alcohol treated for 30 s,followed by rinsed with sterile water for 3 times,then disinfected with 0.1%HgCl_(2) for 6 min and rinsed with sterile water for 5 times;(2)The optimal medium for the proliferation of adventitious buds was WPM+0.5 mg·L^(-1)6-BA+0.1 mg·L^(-1) NAA;(3)The optimal medium for the induction of adventitious root was 1/2WPM+0.3 mg·L^(-1) IBA+0.3 mg·L^(-1) NAA;(4)The survival rate was 100% when the well-grown tissue culture seedlings were transplanted and cultured with peat-loess mixture(mixed in 50/50 ratio by volume).Our study provided technical support for the research on the selection,breeding and genetic transformation of H.tiliaceus.
