摘  要:目的探讨IKKβ及其下游因子IκBα、NF-κBp65在盆腔器官脱垂(pelvic organ prolapse,POP)患者骶韧带中的表达及意义.方法收集POP分度Ⅱ-Ⅳ度患者(POP组、30例),非盆底功能障碍患者(对照组、28例)的子宫骶韧带组织.通过免疫组织化学和Western blot检测IKKβ、IκBα及NF-κBp65的表达.HE染色和Masson染色观察宫骶韧带组织的病理学改变和胶原纤维分布.结果POP组患者子宫骶韧带组织结构疏松紊乱,其胶原纤维数量明显减少.与对照组相比,POP组骶韧带组织中IKKβ、NF-κBp65蛋白表达水平显著增加(P<0.01),而IκBα表达水平明显降低(P<0.01).结论POP患者骶韧带组织中IKKβ及其下游因子表达水平的变化,提示NF-κB信号通路中的关键因子可能参与POP发病.Objective To investigate the expression and significance of IKKβand its downstream factors IκBαand NF-κBp65 in the sacral ligament of patients with pelvic organ prolapse(POP).Methods The uterosacral ligament tissues of patients with POP GradesⅡ-Ⅳ(POP group,30 cases)and non-pelvic floor dysfunction patients(control group,28 cases)were collected.The expressions of IKKβ,IκBαand NF-κBp65 were assessed by immunohistochemistry and Western blot.HE staining and Masson′s trichrome staining were used to observe the pathological changes and the distribution of collagen fibers in the uterine sacral ligament.Results In POP group,the structure of uterosacral ligament was loose,and the number of collagen fibers was significantly decreased.Compared with the control group,the protein expression levels of IKKβand NF-κBp65 in the sacral ligament tissue of the POP group were significantly increased,while the expression level of IκBαwas significantly reduced(P<0.01).Conclusion The changes in the expression levels of IKKβand its downstream factors in the sacral ligament tissue of patients with POP suggest that the key factors in NF-κB signaling pathway may be correlated to the pathogenesis of POP.
