摘 要:目的 分析活化T细胞核因子2(NFAT2)和线粒体外膜电压依赖性阴离子通道蛋白(VDAC)在川崎病(KD)患儿外周血中性粒细胞中的表达以及其相关性。方法 61例住院KD患儿(KD组)中,29例有冠状动脉(冠脉)损害(KD-CAL组),32例无冠脉损害(KD-NCAL组)。采集KD组、46例呼吸道感染伴发热患儿(发热对照组)和58例健康体检儿童(正常对照组)的外周静脉血,提取中性粒细胞,采用实时荧光定量PCR检测中性粒细胞中NFAT2和VDAC mRNA表达,分析NFAT2 mRNA与VDAC mRNA表达的相关性。结果 治疗前,KD组NFAT2、VDAC1、VDAC2和VDAC3 mRNA表达均低于正常对照组和发热对照组(P<0.05),KD-CAL组和KD-NCAL组NFAT2、VDAC1、VDAC2和VDAC3 mRNA表达差异无统计学意义(P>0.05)。治疗后,KD组NFAT2、VDAC1、VDAC2及VDAC3 mRNA表达均高于治疗前(P<0.05),且与正常对照组及发热对照组比较差异无统计学意义(P>0.05)。KD组NFAT2 mRNA与VDAC1、VDAC2、VDAC3 mRNA表达均呈正相关(rs=0.481、0.567、0.479,P<0.01)。结论 NFAT2可能靶向VDAC的表达异常是KD中性粒细胞发生异常的原因之一。Objective To analyze the expressions and correlation of nuclear factor 2of activating T cells(NFAT2)and mitochondrial outer membrane voltage-dependent anion channel protein(VDAC)in peripheral blood neutrophils of the children with Kawasaki disease(KD).Methods Of 61hospitalized children with KD(group KD),29cases had coronary artery lesion(group KD-CAL)and 32cases had no coronary artery lesion(group KD-NCAL).The peripheral venous blood of group KD,46children with respiratory tract infection and fever(group F)and 58healthy children(group C)was drawn to extract neutrophils.The expressions of NFAT2and VDAC mRNA in neutrophils were detected by quantitative real-time PCR.The correlation between NFAT2mRNA and VDAC mRNA expressions was analyzed.Results Before treatment,the expressions of NFAT2,VDAC1,VDAC2and VDAC3 mRNA in group KD were lower than those in groups of F and C(P<0.05),which were not obviously different between group KD-CAL and group KD-NCAL(P>0.05).After treatment,the expressions of NFAT2,VDAC1,VDAC2and VDAC3 mRNA in group KD were higher than those before treatment(P<0.05),which were not obviously different among groups of KD,F and C(P>0.05).The expression of NFAT2mRNA was positively correlated with the expressions of VDAC1,VDAC2and VDAC3mRNA in group KD(rs=0.481,0.567,0.479,P<0.01).Conclusion Abnormal expressions of NFAT2and targeted VDAC may be involved in the neutrophilic abnormality in KD.
