摘 要:为建立快速溶剂萃取–高效液相色谱–三重四极杆串联质谱(ASE-HPLC-MS/MS)检测以肝、肾组织为代表的生物样本中MDMB-4en-PINACA及其代谢物的方法,本文考察了液液萃取、快速溶剂萃取对生物样本中药物的提取效果,用高效液相色谱-三重四极杆串联质谱对生物样本中MDMB-4en-PINACA及代谢物进行分析。结果显示,快速溶剂萃取法回收率最高,在0.5~200 ng/g范围内生物样本中MDMB-4en-PINACA、其水解产物(M1)和脱烷烯基代谢物(M2)浓度与峰面积线性关系良好(r大于0.997 3),MDMB-4enPINACA、M1和M2在肝组织中最低检出限均为0.01 ng/g;MDMB-4en-PINACA、M1和M2在肾组织中最低检出限分别为0.01、0.03和0.02 ng/g。快速溶剂萃取–液相色谱-串联质谱法操作简单、回收率高,可用于生物样本中MDMB-4en-PINACA及其代谢物的检验。To establish an accelerated solvent extraction liquid chromatography-tandem mass spectrometry(ASE-HPLC-MS/MS)method for detecting MDMB-4en-PINACA and its metabolites in biological samples,the effects of liquid-liquid extraction and accelerated solvent extraction on drugs in liver and kidney tissue were investigated.Then the MDMB-4en-PINACA and its metabolites in liver and kidney tissue were analyzed by HPLC-MS/MS.The results showed the ASE had the best performance with a good linear relationship of the MDMB-4en-PINACA,and its hydrolytic metabolite(M1)and dealkenyl metabolite(M2)with the peak area(r>0.9973)in liver and kidney tissue within the range of 0.5–200 ng/g.The LOD values of MDMB-4en-PINACA,M1 and M2 were all 0.01 ng/g in liver tissue.The lower limit of detection values of MDMB-4en-PINACA,M1 and M2 were 0.01,0.03 and 0.02 ng/g in kidney tissue,respectively.The method has the advantages of easy operation and high recovery.It could be concluded that theASE-HPLC-MS/MS can be used to detect the MBMD-4en-PINACAand its metabolites in biological samples.
