摘 要:目的:优化新疆鼠尾草根萜类成分的提取纯化工艺。方法:建立能同时测定新疆鼠尾草根中6,7-去氢罗列酮、齐墩果酸和熊果酸3个萜类成分质量分数的HPLC方法,以3个成分的质量分数为指标,在单因素试验基础上,用Box-Behnken响应面法设计分析分别考察提取温度、提取时间和料液比3个因素对萜类成分提取工艺的影响;考察萃取溶剂比例、萃取次数和振摇时间3个因素对萜类成分纯化工艺的影响。结果:最佳提取工艺为加20倍95%乙醇,85℃回流提取3次,每次2.5 h,在此条件下测得3个成分质量分数之和为4.13%;最佳纯化工艺为向提取物的水混悬液中加等比例的乙酸乙酯萃取5次,每次振摇24 h,在此条件下测得3个成分质量分数之和为21.53%,是提取物的5.19倍。结论:响应面法优选新疆鼠尾草根中萜类成分的提取纯化工艺合理,可为制备新疆鼠尾草根萜类成分提供参考,也为进一步开发利用新疆鼠尾草提供科学依据。OBJECTIVE To optimize the extraction and purification process of terpenoids from roots of Salvia deserta Schang.METHODS To establish a high performance liquid chromatography(HPLC)method for the determination of three constituents,6,7-dehydroroyleanone,oleanolic acid and ursolic acid in roots of Salvia deserta Schang.By taking the mass fraction of the three constituents as the index,the Box-behnken response surface method was used to design and analyze and to optimize the extraction process based on the results of single factor experiment on the extraction temperature,extraction time and solid-liquid ratio.And the purification process was optimized based on the results of single factor experiment on the extraction solvent ratio,extraction times and vibration times by the Box-behnken response surface method.RESULTS The optimal extraction conditions were as follows:20 times of 95%enthanol,refluxing and extracting for 2.5 hat 85℃,totally extracting 3 times.Under these conditions,the mass fraction of three terpenoids was 4.13%.The optimal purification conditions were to add equal proportion of ethyl acetate to the aqueous suspension of the extract for 5 times and shake 24 h each time.Under these conditions,the mass fraction of three terpenoids was 21.53%,which was 5.19 times higher than that before purification.CONCLUSION By using response surface method to optimize extraction and purification process of terpenoids from roots of Salvia deserta Schang is reasonable,which provides a reference for the preparation of terpenoids and scientific basis for the further development and utilization of Salvia deserta Schang.
