摘  要:目的:评价立方液晶载体对5-氨基酮戊酸皮肤渗透性、光动力疗效、稳定性等的影响。方法:以累积渗透量和皮肤滞留量为评价指标,比较5-氨基酮戊酸水溶液(ALA-W)、5-氨基酮戊酸立方液晶纳米凝胶(ALA-CS)的皮肤渗透性差异;以细胞存活率为评价指标,比较ALA-W、ALA-CS的光动力疗效差异;比较ALA-W、ALA-CS的高温稳定性和强光照稳定性。结果:10%ALA-CS的4 h累积渗透量(Q4)和皮肤滞留量显著高于10%ALA-W(P<0.05),但低于20%ALA-W(P<0.05);ALA-CS的光毒性在各个考察浓度均大于ALA-W;ALA-W高温、强光照下不稳定,ALA-CS在高温、强光照下均呈现较好的稳定性。结论:立方液晶载体能有效提高5-氨基酮戊酸的皮肤渗透性、光动力疗效及稳定性,是一种优良的经皮给药制剂载体。OBJECTIVE To evaluate the effect of cubic liquid crystal on skin permeability,photodynamic efficacy and stability of 5-aminolevulinic acid.METHODS The skin permeability of 5-aminolevulinic acid aqueous solution(ALA-W)and 5-aminolevulinic acid cubic liquid crystal nanogels(ALA-CS)were compared by using cumulative permeability andskin retentionas the evaluation indexes.The photodynamic efficacy of ALA-W and ALA-CS was compared using cell viability as the evaluation index.The stability of ALA-W and ALA-CS at high temperature and strong light were studied.RESULTS The 4-hour cumulative permeability(Q4)andskin retentionof 10%ALA-CS were significantly higher than that of 10%ALA-W(P<0.05),while lower than that of 20%ALA-W(P<0.05).ALA-W is unstable under high temperature and strong light,while ALA-CS is stable.CONCLUSION The cubic liquid crystal can effectively improve the skin permeability,photodynamic efficacy and stability of 5-aminophenolvalic acid,which is an excellent carrier for transdermal drug delivery.
