摘  要:山茱萸新苷(cornuside,CNS)是山茱萸(Cornus officinalis Sieb.et Zucc.)中的一种环烯醚萜苷化合物,具有心脏保护作用。为考察山茱萸新苷对力竭训练诱导的大鼠心肌损伤的保护作用,本研究通过力竭游泳建立了力竭训练大鼠模型,训练当天对大鼠分别灌胃3个剂量(10,20和40 mg/kg)的山茱萸新苷,共给药4周。然后检测了大鼠的力竭时间、能量代谢指标、心肌损伤指标、心肌炎症和氧化应激指标。通过Western blot检测心肌组织中Nrf2和NF-κB信号转导途径相关蛋白的表达。结果表明,3种剂量的山茱萸新苷均提高了大鼠的力竭时间,升高了肝糖原、肌糖原和NEFA水平,降低了LA和BUN水平,降低了血清LDH、CK和cTnI水平,降低了心肌组织IL-1β、IL-6和TNF-α水平,升高了心肌组织SOD和CAT水平,降低了MDA水平,升高了心肌组织细胞核Nrf2、HO-1和NQO-1蛋白相对表达量,降低了Keap1和p-NF-κB p65蛋白相对表达量(P<0.05)。山茱萸新苷对上述指标的影响具有剂量依赖性(P<0.05)。本研究表明山茱萸新苷可减轻力竭训练诱导的大鼠心肌损伤,其机制与改善能量代谢、激活Nrf2信号转导途径和失活NF-κB信号转导途径有关。Cornuside(CNS)is an iridoid glycoside compound in Cornus officinalis Sieb.et Zucc..which has cardioprotective effects.The purpose of this study was to investigate the protective effect of cornuside on myocardial injury induced by exhaustive training in rats.In this study,the rat model of exhaustive training was established by exhaustive swimming.On the training day,the rats were given three doses of cornuside(10,20 and 40 mg/kg)for 4 weeks.Then the exhaustion time,energy metabolism index,myocardial injury index,myocardial inflammation and oxidative stress index were measured.The expression of Nrf2 and NF-κB signal transduction pathway related proteins in myocardial tissue was detected byWestern blot.The results showed that all three doses of cornuside increased the exhaustion time,increased the levels of liver glycogen,muscle glycogen and NEFA,decreased the levels of LA and BUN,decreased the levels of serum LDH,CK and cTnI,decreased the levels of IL-1β,IL-6 and TNF-αin myocardial tissue,increased the levels of SOD and CAT in myocardial tissue,decreased the level of MDA,increased the relative expression of Nuclear Nrf2,HO-1 and NQO-1 protein in myocardial tissue,and decreased the relative expression of Keap1 and p-NF-κB p65 protein(P<0.05).The effect of cornuside on the above indexes was dose-dependent(P<0.05).This study shows that cornuside can reduce myocardial injury induced by exhaustive training in rats,and its mechanism is related to the improvement of energy metabolism,activation of Nrf2 signal transduction pathway and inactivation of NF-κB signal transduction pathway.
