药用植物甘草活性成分联合有氧运动对肥胖大鼠减脂效果及基因表达的影响  被引量:1

摘  要:药用植物甘草的活性成分是甘草酸(glycyrrhizic acid,GA),属于甘草中的三萜皂苷,不仅具有抗氧化和调节脂代谢作用,而且可作为保健甜味剂。本研究探讨了甘草酸联合有氧运动对肥胖大鼠的减脂效果及脂代谢基因表达的影响。将大鼠分组、建模和实验处理。运动及给药结束后,检测了各组大鼠的血清脂代谢指标(TC,TG,LDL-c,HDL-c,FFA)和肝脏抗氧化指标(SOD,CAT和MDA)水平。通过肝脏HE染色评价肝脏脂肪积累情况。通过RT-qPCR检测肝脏AMPKα、PGC-1α、PPAR-γ、SREBP1和FAS的m RNA水平。通过Western blot检测肝脏AMPKα磷酸化水平。结果显示,与HF+AE组和HF+GA组比较,HF+AE+GA组大鼠的体重降低,血清TC、TG、LDL-c和FFA水平降低,血清HDL-c水平升高,肝脏脂肪滴数量减少,肝脏SOD和CAT水平升高,肝脏MDA水平降低,肝脏AMPKα、PGC-1α和PPAR-γmRNA水平升高,肝脏SREBP1和FAS mRNA水平降低,肝脏AMPKα磷酸化水平升高(P<0.05)。本研究表明,有氧运动联合甘草酸可能通过调控AMPK信号传导途径中的脂代谢基因转录和表达来减少肥胖大鼠脂肪堆积。考虑到甘草酸本身具有的甜味剂属性,本研究认为甘草酸可能是一种有效的运动营养补充剂。The active ingredient of the medicinal plant licorice is glycyrrhizic acid(GA),which belongs to the triterpenoid saponins in licorice.It not only has antioxidant and lipid metabolism regulating effects,but also can be used as a health sweetener.This study investigated the effects of glycyrrhizin combined with aerobic exercise on lipid reduction and gene expression of lipid metabolism in obese rats.In this study,rats were grouped,modeled,and subjected to experimental treatment.After exercise and administration,the levels of serum lipid metabolism indicators(TC,TG,LDL-c,HDL-c,FFA)and liver antioxidant indicators(SOD,CAT,and MDA)were measured in each group of rats.Liver fat accumulation was evaluated by liver HE staining.The mRNA levels of AMPKα,PGC-1α,PPAR-γ,in liver were measured by RT-qPCR.Liver AMPKαphosphorylation levels were measured by Western blot..The results showed that compared with the HF+AE group and the HF+GA group,the weight of the HF+AE+GA group rats decreased,the serum TC,TG,LDL-c,and FFA levels decreased,the serum HDL-c level increased,the number of liver fat droplets decreased,the liver SOD and CAT levels increased,the liver MDA level decreased,and the mRNA levels of AMPKα,PGC-1αand PPAR-γin liver were increase,the SREBP1 and FAS mRNA levels were decrease,AMPKαphosphorylation level in liver was increase(P<0.05).This study suggests that aerobic exercise combined with glycyrrhizin may reduce fat accumulation in obese rats by regulating the transcription and expression of lipid metabolism genes in the AMPK signaling pathway.Considering the sweetener properties of glycyrrhizic acid itself,this study suggests that glycyrrhizic acid may be an effective sports nutrition supplement.
