补肾活血方对精索静脉曲张大鼠生精功能的影响

摘　　要：目的:基于细胞周期调控因子细胞周期蛋白D1(cyclin D1)、细胞周期蛋白依赖激酶4(cdk4)的表达探讨补肾活血方改善精索静脉曲张大鼠生精功能的机制。方法:将30只大鼠随机均分为假手术组,模型组,补肾活血方低、中、高剂量组,每组各6只。建立左侧精索静脉曲张模型,造模成功后补肾活血方低、中、高剂量组分别灌胃补肾活血方药液6、12、24 g/kg·d^(-1),其余2组灌胃10 ml/kg·d^(-1)的0.9%氯化钠注射液,灌胃30 d。检测精子浓度、活力、畸形率及DNA碎片率,HE染色观察睾丸组织形态,免疫组化分析睾丸cyclin D1、cdk4表达。结果:在大鼠浓度、精子活力、精子畸形率、精子DNA碎片率、睾丸病理评分及cyclin D1、cdk4表达方面模型组与假手术组比较,补肾活血方低、中、高剂量组与模型组比较,差异均有统计学意义(P<0.05或P<0.01);补肾活血方低、中、高剂量组精子浓度、活力、畸形率、DNA碎片率组间比较,差异均无统计学意义(P>0.05)。结论:补肾活血方可能通过调控cyclin D1、cdk4表达来改善精索静脉曲张大鼠的精子发生。Objective:To investigate the mechanism of kidney-tonifying and blood-activating prescription improving the spermatogenic function of rats with varicocele based on the expression of the cell cycle regulators cyclin D1 and cyclin-dependent kinase 4(cdk4).Methods:A total of 30 rats were randomly divided into shamoperation group,model group,and low-,middle-,high-dose kidney-tonifying and blood-activating prescription groups,with 6 rats in each group.A model of left varicocele was established;after successful modeling,the rats in the low-,middle-,high-dose groups were given the solution of kidney-tonifying and blood-activating prescription by gavage at a dose of 6,12,and 24 g/kg·d^(-1),respectively,and those in the sham-operation group and the model group were given 0.9%sodium chloride injection by gavage at a dose of 10 ml/kg·d^(-1),for 30 days.The rats were observed in terms of sperm concentration,sperm motility,rate of sperm deformation,and sperm DNA fragmentation rate;HE staining was used to observe the histomorphology of the testis,and immunohistochemistry was used to measure the expression of cyclin D1 and cdk4 in the testis.Results:There were significant differences in sperm concentration,sperm motility,rate of sperm deformation,sperm DNA fragmentation rate,pathological score of the testis,and the expression of cyclin D1 and cdk4 between the model group and the sham-operation group,as well as between the low-,middle-,high-dose groups and the model group(P<0.05 or P<0.01).There were no significant differences in sperm concentration,sperm motility,rate of sperm deformation,and sperm DNA fragmentation rate between the low-,middle-,high-dose groups(P>0.05).Conclusion:Kidney-tonifying and blood-activating prescription can improve the spermatogenic function of rats with varicocele by regulating the expression of cyclin D1 and cdk4.
