摘  要:目的 对月腺大戟(Euphorbia ebracteolata Hayata)根的体积分数95%乙醇提取物中的化学成分进行分离鉴定,并初步评价其抗肿瘤活性。方法 运用硅胶、Sephadex LH-20、反相十八烷基硅烷键合硅胶(ODS)等柱色谱以及反相高效液相色谱方法对月腺大戟化学成分进行分离和纯化,并利用高分辨率质谱(HR-ESI-MS)、核磁共振(NMR)等波谱技术对分离得到的化合物进行结构鉴定;采用CCK-8法测试化合物对人肝癌HepG2细胞,人乳腺癌MCF-7细胞和人肺癌A549细胞的抗肿瘤活性。结果 从月腺大戟根的体积分数95%乙醇提取物中共分离得到12个化合物,分别鉴定为portulacanone A(1)、甘草素(2)、异甘草素(3)、芒柄花素(4)、24-亚甲基环木菠萝烷醇(5)、3β-羟基-9(11), 12-齐墩果二烯(6)、橙皮油醇(7)、(+)-水合橙皮内酯(8)、2, 2′-氧代双(1,4-二叔丁苯)(9)、反-4-羟基-2-壬烯酸(10)、(E)阿魏酸二十六烷酯(11)和1,9-壬二酸(12),并对化合物1~8进行了抗肿瘤活性研究。结果表明,化合物5(24-亚甲基环木菠萝烷醇)对人肺癌细胞A549具有显著的抑制作用,其半数抑制浓度(median inhibition concentration, IC50)为26.03μmol·L^(-1)。结论 化合物1,7~10为首次从大戟属植物中分离得到,化合物1~4和6~12为首次从月腺大戟中分离得到。OBJECTIVE To isolate and identify the chemical constituents in the 95%ethanol extract of roots from Euphorbia ebracteolata Hayata and evaluate their antitumor activity.METHODS The compounds were systematically iaolated and purified by various chromatographic iaolation and analysis techniques including macroporous resins,silica gel,Sephadex LH-20 and ODS column chromatographies,as well as reversed phase high-performance liquid chromatography.The structures of the compounds were mainly identified by HR-ESI-MS and NMR spectral techniques.The antitumor activities of the compounds against human liver cancer HepG2 cells,human breast cancer MCF-7 cells and human lung cancer A549 cells were tested by CCK-8 assay.RESULTS A total of twelve compounds were isolated from the 95%ethanol extract of E.ebracteolata root and identified as portulacanone A(1),liquiritigenin(2),isoliquiritigenin(3),formononet(4),24-methylenecycloar-tanol(5),3β-hydroxyoleana-9(11),12-diene(6),auraptenol(7),(+)-meranzin hydrate(8),2,2′-oxybis(1,4-di-tert-butylbenzene)(9),trans-4-hydroxy-2-nonenoic acid(10),hexacosyl(E)-ferulate(11),and 1,9-azelaic acid(12).The antitumor activity of compounds 1-8 was studied,and the results showed that compound 5 can effectively inhibit the growth of A549 cells,and its median inhibitory concentration(IC50)was 26.03μmol·L^(-1).CONCLUSION Compounds 1,7-10 are found in the genus Euphorbia Linn.for the first time.In addition,compounds 1-4,6-12 are isolated from E.ebracteolata for the first time.
