摘 要:目的观察黄芩素对脑缺血性眩晕大鼠的作用及作用机制。方法建立脑缺血性眩晕大鼠模型,以盐酸氟桂利嗪作为阳性对照,用黄芩素治疗模型大鼠;跳台实验检测大鼠跳台潜伏期;迷宫实验检测大鼠逃避电击时间;试剂盒检测LAC、LDH、MDA和SOD水平;蛋白印记实验检测脑组织GABABR和GAD67蛋白表达变化。结果脑缺血性眩晕大鼠模型构建成功。盐酸氟桂利嗪(0.5 mg/kg和2 mg/kg)和黄芩素(50 mg/kg和200 mg/kg)各剂量组均能缩短缺血性眩晕大鼠的跳台潜伏期和迷宫逃避电击能力(P<0.01),降低脑组织LDH和MDA含量(P<0.01),增加前庭神经核组织血流量(P<0.01)。黄芩素(200 mg/kg)还能降低脑缺血性眩晕大鼠LAC和SOD含量(P<0.01),且上调脑组织GABABR和GAD67蛋白表达(P<0.01)。结论黄芩素对脑缺血性眩晕大鼠具有治疗作用,该作用可能与增加前庭核血流量、降低氧化损伤及上调脑组织GABABR和GAD67蛋白表达有关。Objective To observe the effect and mechanism of baicalein on rats with cerebral ischemic vertigo.Methods A rat model of cerebral ischemic vertigo was established.With flunarizine hydrochloride as a positive control,model rats were treated with baicalein.The platform jumping test was used to detect the platform jumping latency of rats.The maze test was used to detect the escape time of rats to electric shock.The kit was used to detect LAC,LDH,MDA and SOD levels.The protein expression changes of GABABR and GAD67 in brain tissue were detected by western blotting.Results The rat model of cerebral ischemic vertigo was successfully constructed.Flunarizine hydrochloride(0.5 and 2 mg/kg)and baicalein(50 and 200 mg/kg)in each dose group could shorten the platform jump latency and maze escape ability in ischemic vertigo rats,with statistical significance(P<0.01).The contents of LDH and MDA in brain tissue were decreased(P<0.01),and the blood flow of vestibular nerve nucleus was increased,with statistical significance(P<0.01).Baicalein(200 mg/kg)could also reduce the contents of LAC and SOD in rats with cerebral ischemic vertigo(P<0.01),and up-regulate the protein expressions of GABABR and GAD67 in brain tissue,with statistical significance(P<0.01).Conclusion Baicalein has a therapeutic effect on ischemic vertigo rats,which may be related to increasing vestibular nuclear blood flow,reducing oxidative stress and up-regulating GABABR and GAD67 protein expression in brain tissue.
