摘 要:目的观察加味八正散对慢性肾小球肾炎模型大鼠血清肝细胞生长因子(HGF)及血管内皮生长因子(VEGF)的影响,探讨加味八正散治疗慢性肾小球肾炎的作用机制。方法将90只SD大鼠适应性饲养7 d后,采用随机数字表法分为空白组,模型组,阳性对照组,加味八正散低、中、高剂量组6组,每组15只。除空白组外,其余各组大鼠均在腹股沟处皮下及尾静脉注射阳离子化牛血清白蛋白溶液,建立慢性肾小球肾炎大鼠模型。造模成功后,加味八正散高、中、低剂量组分别灌胃加味八正散溶液36,18,9 g/(kg·d),阳性对照组灌胃肾炎康复片混悬液0.6 mg/(kg·d),空白组与模型组均灌胃相同体积的蒸馏水,连续灌胃6周。末次给药后,大鼠腹主动脉取血,分离血清,采用酶联免疫吸附法测定各组大鼠血清HGF、VEGF含量。结果与空白组比较,模型组大鼠血清HGF、VEGF含量明显降低(P<0.01);与模型组比较,加味八正散各剂量组及阳性对照组大鼠血清HGF、VEGF含量均明显升高(P<0.01)。结论加味八正散可升高慢性肾小球肾炎模型大鼠血清HGF、VEGF含量,促进肾脏间质血管再生和修复。Objective To observe the effect of modified Bazheng San(八正散)on serum hepatocyte growth factor(HGF) and vascular endothelial growth factor(VEGF) in chronic glomerulonephritis model rats, and to explore the mechanism of action of modified Bazheng San in treating chronic glomerulonephritis.Methods After adaptive feeding for 7 days, 90 SD rats were randomly divided into 6 groups using a random number table method: a blank group, a model group, a positive control group, and low, medium, and high dose groups of modified Bazheng San, with 15 rats in each group. Except for the blank group, all other groups of rats were subcutaneously injected with cationic bovine serum protein solution in the groin and caudal vein to establish the rats model of chronic glomerulonephritis. After successful modeling, the high, medium, and low dose groups of modified Bazheng San were gavaged with 36,18,and 9 g/(kg·d) of modified Bazheng San solution, the positive control group was gavaged with 0.6 mg/(kg·d) of Shenyan Kangfu Pian(肾炎康复片)suspension, and the blank group and model group were gavaged with the same volume of distilled water for 6 consecutive weeks. After the last administration, blood was taken from the abdominal aorta of rats, and serum was separated. The levels of serum HGF and VEGF in each group of rats were measured using enzyme-linked immunosorbent assay. Results Compared with the blank group, the content of serum HGF and VEGF in the model group rats decreased(P<0.01). Compared with the model group, the serum HGF and VEGF levels of rats in each dose group of modified Bazheng San and the positive control group were significantly increased(P<0.01). Conclusion Modified Bazheng San can increase the levels of serum HGF and VEGF in chronic glomerulonephritis model rats, promote renal interstitial blood vessels of kidney regeneration and repair.
